Role of arginine and its metabolism in physiology of macrophages

Abstract:

Bylo prokázáno, že arginin má různorodé a mnohdy protikladné efekty v rámci různých buněčných populací, včetně makrofágů, které mohou být způsobeny změnou v regulaci vni-trobuněčných drah. Cílem této disertační práce bylo charakterizovat molekulární mecha-nismy, zahrnuty v regulaci makrofágů zprostředkované argininem a objasnit zda změna mi-mobuněčné dostupnosti argininu hraje roli v produkci superoxidového aniontu a oxidu dus-natého indukované lipopolysaccharidem. Ze získaných dat vyplývá, že arginin reguluje pro-dukci oxidu dusnatého u makrofágů minimálně dvěma způsoby: (1) regulací jeho dostup-nosti pro inducibilní syntasu oxidu dusnatého, (2) a regulací aktivity mitogenen ak-tivovaných protein kináz/nukleárního faktoru-kappaB, která je dále doprovázená redukcí exprese induciblní syntasy oxidu dusnatého. Navíc se ukázalo, že snížení exprese inducibilní syntasy oxidu dusnatého je spojeno s redukcí produkce superoxidového aniontu u makrofágů dlouhodobě stimulovaných pomocí lipopolysaccharidu. Naše výsledky naznačují, že NADPH oxidasa spolu s inducibilní syntasou oxidu dusnatého může spůsobit tzv. „un-coupling“, který vede ke zvýšení produkce superoxidového aniontu a také k tvorbě vysoce reaktivního peroxynitritu. Předpokládáme, že ovlivnění exprese inducibilní syntasy oxidu dusnatého, produkce superoxidového aniontu a oxidu dusnatého mimobuněčným argininem hraje důležitou roli v zánětlivých odpovědích makrofágů. Navíc naše data naznačují, že regu-lace zmíněných intracelulárních drah argininem může být zprostředkovaná jeho přímou in-terakcí s povrchovými receptory, které ovlivňují klíčové funkce makrofágů.

Abstract:

Arginine exerts the pleiotropic and often contrasting effects on different cell populations in-cluding macrophages which can be mediated by a number of factors that are still mostly ei-ther undefined or speculative. The aim of this Ph.D. Thesis was to characterize molecular mechanisms involved in arginine-mediated regulation of macrophage physiology and to set-tle the controversy of whether the deregulation of the extracellular arginine availability play a key role in LPS-derived macrophage superoxide anion and nitric oxide production. Our data demonstrated that the impact of arginine on the regulation of inducible nitric oxide synthase-mediated NO synthesis is due to two different effects: (1) lowered substrate availability for inducible nitric oxide synthase and (2) diminished mitogen activated protein kinases/nuclear factor kappaB activity which is followed by decreased inducible nitric oxide synthase protein expression. Furthermore, we demonstrated that down regulation of inducible nitric oxide synthase protein expression is directly associated with the reduction of superoxide anion production in the long-term incubated macrophages stimulated with bacterial endotoxin. Our results indicate that simultaneous activation of NADPH oxidase together with induction of inducible nitric oxide synthase can result to uncoupled state of inducible nitric oxide synthase early after macrophage activation with lipopolysaccharide and thus can reduce the produc-tion of nitric oxide and cause increase in superoxide anion and peroxynitrite formation. We suppose that the expression and regulation of inducible nitric oxide synthase protein expres-sion, as well as superoxide anion and nitric oxide production in macrophages by removal of arginine or prevention of its availability could have an important role in the inflammatory response of macrophages. Furthermore, it can be hypothesized that arginine may be directly associated with the regulation of common receptor-dependent intracellular pathways which affect critical macrophage functions.
