The effect of physical factors on photosynthesis in extremophillous organisms

Abstract:

Tato diplomová práce je zaměřena na vliv fyzikálních faktorů na fyziologii fotosyntézy modelových druhů extremofilních řas z Antarktidy. Pro studium fyziologických charakteristik modelových druhů byly zvoleny 2 specifické experimenty. Experiment A byl zaměřen na vliv různých úrovní ozářeností při teplotním gradientu na jejich fotosyntézu. Testovanými druhy byly Klebsormidium sp. a Trebouxia sp. Experiment B byl zaměřen na projevy repetitivní fotoinhibice na primární fotochemické procesy fotosyntézy a testovanými druhy byly Chlamydomonas reinhardtii, Klebsormidium sp. a Trebouxia sp. U obou experimentů bylo pro hodnocení fyziologického stavu využito indukované fluorescence chlorofylu, sledovaným parametrem byl efektivní kvantový výtěžek fotosyntetických procesů ve fotosystému II (ΦII). U experimentu A bylo pro hodnocení fyziologického stavu navíc využito měření rychlosti vývinu kyslíku (OER) a při experimentu B byly dále sledovány změny v množství biomasy, celkový obsah Chl a, Chl b a celkový obsah karotenoidů v průběhu kultivace obou druhů ve fotobioreaktoru.

Abstract:

This diploma thesis is focused on the impact of physical factors to physiology of photosynthesis of model species of extremophilic algae from Antarctica. For study of physiologic characteristics of model species ,two specific experiments were carried out. Experiment A was focused on the impact of different levels of photosynthetically active radiation in temperature gradient to photosynthesis of two model species. Tested species were Klebsormidium sp. and Trebouxia sp. Experiment B was focused on the effects of repetitive photoinhibition on primary processes of photosynthesis. In this experiment, Chlamydomonas reinhardtii, Klebsormidium sp. a Trebouxia sp. were tested species. In both experiments, induced fluorescence of chlorophyll was used for the evaluation of their physiological state. Efective quantum yield of photosynthetic processes in photosystem II (ΦII) was used as a parameter indicating changes in photosynthesis. In experiment A, measurements of rate of oxygen evolution (OER) was used for evaluation of physiological state. Additionally, changes in amount of biomass and total contain of Chl a, Chl b and total carotenoids during cultivation of all of three model species in photobioreactor were evaluated in experiment B.
