Identification of bacteria of genus Bifidobacterium using DNA amplification methods

Abstract:

Bakterie rodu Bifidobacterium tvoří jednu z největších bakteriálních populací v lidském trávicím traktu. Tyto bakterie jsou Gram-pozitivní, nepohyblivé, nesporulující, anaerobní tyčinky. Pro druhovou identifikaci bakterií rodu Bifidobacterium byly použity molekulárně biologické metody. Templátová DNA byla získána z 41 kmenů bifidobakterií, které byly izolovány ze stolice zdravých, plně kojených dětí. Pro ověření správnosti zařazení kmenů do rodu Bifidobacterium byla nejprve provedena PCR s rodově specifickými primery. Následně byla provedena druhová identifikace pomocí PCR s druhově specifickými primery a na závěr byla se všemi vzorky provedena metoda rep-PCR.

Abstract:

One of the largest bacterial populations in the human gastrointestinal tract is formed by bifidobacteria. All members of the genus Bifidobacterium are Gram-positive, non-motile, non-sporulating, anaerobic microorganisms. In order to clarify the distribution of bifidobacterial species in the human intestinal tract, molecular methods were used for the analysis of DNAs isolated from faeces of 41 healthy, breast-fed infants. At first we used genus-specific PCR. Consequently, the species-specific PCR technique was used to identify the Bifidobacterium strains. Finally, the DNA extracts from infant fecal samples were analysed by rep-PCR.
