Bacteriophage lytic enzymes and their applications

Abstract:

U většiny fágů je lyze hostitelské buňky zprostředkována pomocí lytických enzymů, tzv. lyzinů. Mezi tyto lyziny jsou řazeny dva enzymy, které se nazývají holiny a endolyziny. Oba dva tyto lytické enzymy spolu musí spolupracovat, aby dokázaly lyzi navodit. Holiny nejprve zpřístupni endolyzinům peptidoglykan tím, že vytvoří póry v cytoplazmatické membráně. Endolyziny se tak dostanou k peptidoglykanu, kde štěpí jednu ze základních vazeb, čímž narušují integritu buněčné stěny. Když dojde k takovému rozrušení buněčné stěny, tak působením vysokého vnitřního tlaku bakteriální buňky dojde k vytlačování cytoplazmy a k tzv. hypotonické lyzi. Lyze bakteriální buňky pomocí fágových lytických enzymů je velmi účinná metoda, o které se proto začalo uvažovat jako o metodě využitelné pro člověka. Využitelnost fágů a také lytických enzymů jako antibakteriálního agens je například v medicíně, kde v současnosti narůstá rezistence bakterií vůči stále většímu počtu antibiotik. Další oblasti, kde se fágů a jejich lytických enzymů dá využít je veterinární lékařství a také potravinářství, kde je nutno kontrolovat kontaminace potravin různými patogeny. V neposlední řadě se díky specifické vazbě fágů na bakterie mohou fágy využívat k detekci některých bakterií. Perspektivní je také výzkum interakce fágů a lytických enzymů s bakteriálními biofilmy odolnými vůči mnoha dosud objeveným antibiotikům.

Abstract:

Bacteriophage lytic enzymes mediate lysis of the host bacterial cell. The lytic enzymes include two proteins, which are called holins and endolysins. Both of these proteins must work together to be able to induce lysis. Holins first make peptidoglycan accessible for endolysins by forming pores to the cytoplasmic membrane. Endolysins then get to the peptidoglycan, where they cleave one of the basic linkages, thereby disturb the integrity of the cell wall. When the cell wall is disturbed, the effect of high internal pressure of bacterial cell then make the cytoplasm to be expelled. This phenomenon is called the hypotonic lysis. Lysis of bacterial cells using phage lytic enzymes is very effective method, which therefore became thought to be a useful method for human. Uselfulnes of phages and also their lytic enzymes as antibacterial agents is for example in medicine, where it offers a possible alternative to conventional antibiotics in the current era of multidrug resistant pathogens. Another area where phages and their lytic enzymes can be used is veterinary medicine and food industry, where it is necessary to control food contamination by various pathogens. Last but not least, thanks to the specific binding of phages to bacteria, they can be used for detection of certain bacteria.
