Mechanisms taking role in early and late phase of plant defence reaction

Abstract:

Během svého života rostlina musí čelit neustálým vlivům prostředí. Tyto vlivy mohou být rozděleny na biotické a abiotické. V průběhu evoluce si rostliny vyvinuly důmyslné mechanismy, jak se s těmito podmínkami vyrovnat. V reakci na okolí jsou rostliny schopny spustit tzv. obrannou reakci, která je nástrojem pro boj s patogeny i abiotickým stresem jako je například vítr. Rostliny během obranné reakce syntetizují velké množství specifických látek. Pro ranou fázi je typická produkce reaktivních kyslíkových radikálů (ROS). Charakteristickým znakem pozdní fáze obranné reakce rostlin je produkce fytohormonů, fytoalexinů a PR-proteinů. Diplomová práce se z části zaměřila na studium působení biotického a abiotického stresu na suspenzní kultury Révy vinné (Vitis vinifera cv. Gamay). Biotický stres byl reprezentován Plísní šedou (B. cinerea), která je v České republice jedním z nejčastějších patogenů, napadajících révu vinnou. Jako abiotický stresový faktor byl použit mechanický stres. Po působení stresu byly sledovány transkripty genů PR-proteinů, konkrétně PR-10 a chitinas (PR-3 a PR-4). Pomocí qPCR byla sledována míra transkripce u 12 genů pro chitinasy a 10 genů pro PR-10. Nárůst transkripce sledovaných genů chitinas i PR-10 byl zaznamenán po působení bioticého stresu. Mechanický stres však nevyvolal větší změny v transripci chitinas ani PR-10. Druhá část diplomové práce se zaměřila na produkci ROS. V rámci studia rané fáze obranné reakce byla vytvořena nová buněčná linie BY2 (bright yellow) buněk tabáku, exprimující mutantní protein Eos-NtRBOHD. Protein NtRBOHD je pravděpodobně hlavním producentem ROS v buňkách tabáku po elicitaci kryptogeinem. Připojení fluorescenčního proteinu Eos pomůže v budoucnu určit lokalizaci NtRBOHD v buňce během obranné reakce.

Abstract:

Throughout their life, plants must cope with numerous different environmental constraints. They thus have evolved specific mechanisms to overcome such a situation. Among them are the so-called defence reactions, which are powerfully used for counteracting both biotic (pathogens) and abiotic (drought, wind) stresses. A plethora of distinct chemical substances can be synthetized by plants during plant defence reaction. For example, massive production of ROS (reactive oxygen species) is a hallmark of early stages of these responses. Characteristic substances for late stages are phytohormones, phytoalexins and PR-proteins. Diploma thesis was focused in part on the effects of both biotic and abiotic stresses on cell suspensions from the grapevine Vitis vinifera cv. Gamay. The fungus Botrytis cinerea represented the biotic stress. This fungus is one of the most commonly pathogen found to infect V. vinifera in Czech Republic. Mechanical stress was also used as an abiotic stress factor. Transcripts of both chitinases and PR-10 genes were analysed. Transcription levels of 12 chitinases genes and 10 PR-10 genes were quantified by qPCR method. We observed an up-regulation of the transcription of both chitinases and PR-10 after biotic stress. By contrast, mechanical stress did not induce any significant change in the transcript level of any studied genes. The second part of diploma thesis deals with the production of ROS during early stages of plant defence reaction. For this study, transgenic BY-2 (Nicotiana tabaccum) cell lines expressing the mutant protein Eos-NtRBOHD were engineered. The protein NtRBOHD is the likely main producer of ROS after elicitation by cryptogein in tobbaco cells. Fusion of fluorescent protein Eos and NtRBOHD should help to determine dynamics of NtRBOHD in vivo upon elicitation.
