Spectrophotometric determination of thermodynamic dissociation constants of some selected drugs

Abstract:

Spetrofotometrickou titrací s UV detekcí byly stanoveny termodynamické disociační konstanty léčiv butorphanolu tartrát, zolpidemu tartrát a capecitabinu při teplotě 25C a 37C. Na základě Kankareho-Wermontovy faktorové analýzy a regresní analýzy absorbanční matice programem SQUAD(84) byl určen chemický model pro dané léčivo a určen počet světlo-absorbujících částic. Smíšené disociační konstanty byly stanoveny regresní analýzou absorbanční respozní plochy programy SQUAD(84) a SPECFIT32 a následně ze závislosti smíšených disociačních konstant pKa na odmocnině z iontové síly byla při dané teplotě odhadnuta regresní analýzou limitního Debye-Hűckelova vztahu termodynamická disociační konstanta pKTa potenciometrickou titrací byly stanoveny termodynamické disociační konstanty pravastatinu sodného při teplotě 25C. Na základě těsnosti proložení byly stanoveny smíšené disociační konstanty, a to regresní analýzou naměřených dat programem ESAB a HYPERQUAD. Ze závislosti smíšených disociačních konstant pKa na odmocnině z iontové síly byly při teplotě 25C odhadnuty regresní analýzou limitního Debye-Hűckelova vztahu termodynamické disociační konstanty.

Abstract:

Thermodynamic dissociation constants of butorphanoli tartrate, zolpidemi tartrate and capecitabine were estimated by the technique of pH-spectrophotometric titration at 25C a 37C. With the use of Kankare-Wermont factor analysis and program SQUAD(84) was define chemical model of drug. On the basis of a knowledge of the chemical model the number of light-absorbing species were estimated. The mixed-dissociation constants were estimated by the regression analysis of the absorbance response surface using programs SQUAD(84) and SPECFIT32 and consequently from a dependence of dissociation constants on the root of the ionic strength by non-linear regression analysis of Debye - Hückel law the termodynamic dissociation constant pKTa. was estimated. Thermodynamic dissociation constant of pravastatin sodium were estimated by potentiometric titration at 25C. On the basis of a goodness-of-fit there were estimated mixed dissociation constats by the regression analysis of potentiometric data using program ESAB and HYPERQUAD. From dependence of dissociation constants on the root of the ionic strength was by non-linear regression analysis of Debye - Hückel law, the termodynamic dissociation constant pKTa at 25C was estimated.
