Health risks of N-nitrosamines occurring in active pharmacological substances compared to risks associated with other sources of these substances

Abstract:

V úvodní části předložené bakalářské práce jsou krátce shrnuty nejvýznamnější toxické účinky N-nitrosaminů a popsány hlavní zdroje těchto látek ve farmakologicky aktivních ingrediencích a ve finálních léčivých přípravcích. Dále jsou zde shrnuty nejvýznamnější případy léčiv kontaminovaných N-nitrosaminy, které byly zachyceny kontrolními orgány v Evropské unii a USA: Zdravotní rizika představovaná takto kontaminovanými léčivy jsou následně porovnána se zdravotními riziky N-nitrosaminů přítomných v potravinách. K danému účelu byla použita data Evropského úřadu pro bezpečnost potravin. Bylo zjištěno, že nezávisle na typu expozičního scénáře mohou kontaminovaná léčiva podstatným způsobem přispívat ke zdravotním rizikům spojeným s expozicí N-nitrosaminům.

Abstract:

In the introductory part of the presented bachelor thesis, the most important toxic effects of N-nitrosamines are briefly summarized, the main sources of these substances in pharmacologically active ingredients and in final pharmaceuticals are defined. Furthermore, the most important cases of N-nitrosamine-contaminated pharmaceuticals that have been intercepted by regulatory authorities in the European Union and the USA are summarised: the health risks described by such contaminated pharmaceuticals are then compared with the health risks of N-nitrosamines present in food. Data from the European Food Safety Authority were used for this purpose. It was found that, irrespective of the type of exposure scenario, contaminated pharmaceuticals can contribute substantially to health risks associated with exposure to N-nitrosamines.
