Analysis of gluten proteins by the SDS-PAGE method in selected samples of flour

Abstract:

Bakalářská práce pojednává o obecné charakteristice mouk a v nich obsažených proteinů. Zaměřuje se na zásobní proteiny, které zároveň představují proteiny tvořící lepek. Pro analýzu těchto proteinů byla použita metoda separace pomocí elektroforézy v polyakrylamidovém gelu za přítomnosti SDS. Z analýzy vyplývá rozdílné zastoupení frakcí proteinů lepku mezi vybranými vzorky. Z výsledků vyplývá, že hrubost mletí v rámci vybraných vzorků obsah proteinů lepku neovlivňuje. Rozdíly v rámci stejného druhu mouky byly zjištěny často, může se pravděpodobně jednat o důsledek jiné odrůdy obiloviny či k tomuto jevu dochází vlivem vnějších podmínek působících na rostoucí rostlinu. Nejčastěji byly rozdíly zjištěny v gliadinové frakci proteinů. Hlavní rozdíl v gliadinech byl pozorován u špaldové mouky, která na rozdíl od pšeničné mouky, neobsahovala ?-1,2-gliadiny. Vzorek rýžové mouky prokazatelné neobsahoval proteiny lepku vyvolávající autoimunitní odpověď u celiaků a alergiků.

Abstract:

The bachelor's thesis discusses the general characteristics of flours and the proteins contained in them. It focuses on storage proteins, which are also gluten-forming proteins. For the analysis of these proteins, the method of separation using polyacrylamide gel electrophoresis in the presence of SDS was used. The analysis shows a different representation of gluten protein fractions between the selected samples. The results show that the coarseness of grinding within the selected samples does not affect the gluten protein content. Differences within the same type of flour were often found, it can probably be the result of a different variety of cereal, or this phenomenon occurs due to external conditions affecting the growing plant. Most often, differences were found in the gliadin fraction of proteins. The main difference in gliadins was observed in spelled flour, which, unlike wheat flour, did not contain ?-1,2-gliadins. The rice flour sample was demonstrably free of gluten proteins causing an autoimmune response in celiacs and allergy sufferers.
