Lectins from Aspergillus fumigatus and oprimisation of eucaryotic systems for their expression

Abstract:

A. fumigatus je významným alergenem a také patogenem, obzvláště pro lidi s oslabeným imunitním systémem. Jedná se o saprofytickou houbu, která je díky malým vzduchem přenášeným konidím všudypřítomná. Zásadní roli při adhezi konidií na buňky hostitele mohou mít lektiny, což jsou proteiny nebo glykoproteiny specificky vázající sacharidové struktury. Charakterizace lektinů tohoto patogenu a jejich vazebných schopností by mohla být důležitým poznatkem pro vývoj antiadhezivní terapie jako možnosti léčby. Tato práce navazuje na dříve provedené experimenty s lektiny AFL3 a AFL3b z A. fumigatus. Protože exprese v prokaryotním systému E. coli byla obtížná, bylo žádoucí otestovat možnosti exprese v eukaryotním systému. Nejdříve tedy byly dříve připravené konstrukty ověřeny a následně použity pro transformaci P. pastoris GS 115. Exprese AFL3 do média byla potvrzena MS analýzou. Také bylo potřeba navrhnout možnosti purifikace těchto lektinů. Proto byly připraveny konstrukty s oligohistidinovou kotvou na C-konci proteinu, která by umožnila purifikaci pomocí metalo-afinitní chromatografie.

Abstract:

A. fumigatus is an important allergen and also a pathogen especially for immunocompromised people. It is a saprophytic fungus, which is ubiquitous owning to small airborn conidia. Lectins, which are proteins or glycoproteins specifically binding to the sacharide structures, can play a crucial role for the adhesion of conidia to the host cell. Characterization of lectins of this patogen, as well as their binding abilities, can provide important knowledge for antiadhesion therapy development as a possibility for treatment. This thesis follows the previous experiments with AFL3 and AFL3b lectins from A. fumigatus. Due to the difficulties with expression in prokaryotic system E. coli, it was necessary to test the expression in eukaryotic system. Firs, the constructs prepared beforehand were verified and subsequently used for transformation of P. pastoris GS 115. Expression of AFL3 into the medium was confirmed by MS analysis. It was also necessary to propound options for purification of these lectins. Therefore the new constructs were created, with oligohistidine tag at C-terminus of protein, which would facilitate purification by metal – affinity chromatography.
