Vliv abiotického stresu na stárnutí listů v modelových rostlinách Arabidopsis thaliana a Nicotiana tabacum

Leaf senescence in Arabidopsis thaliana and Nicotiana tabacum model plants under abiotic stress conditions

Abstract:

V této diplomové práci se věnujeme reakci rostlin na abiotický stres vyvolaný kadmiem. Tento toxický kov má nepříznivý vliv na růst a vývoj rostlin, a tudíž na celou zemědělskou rostlinou produkci. Stres vyvolaný kadmiem ovlivňuje v rostlinách základní metabolické dráhy a může vést k předčasnému stárnutí. Senescence listů je regulovaná posloupnost katabolických procesů, které vedou k buněčné smrti. Stárnutí listů je spojeno s rozsáhlými změnami v expresi genů. Cílem práce bylo porovnat vliv dvou různých kademnatých solí na růst kořenů klíčních rostlin Arabidopsis thaliana a Nicotiana tabacum a analyzovat hladiny transkriptů námi vybraných genů účastnících se procesů senescence (degradace chlorofylu – ABF4 a NYE1 u rostliny Arabidopsis thaliana; NYC1 a PAO u rostliny Nicotiana tabacum a degradace proteinů – UBC1 a RPT1A u Arabidopsis thaliana; RPT1A u Nicotiana tabacum). Dále byly studován vliv dlouhodobého působení stresu u hydroponicky pěstovaných rostlin Arabidopsis thaliana a Nicotiana tabacum vystavených dvěma environmentálně relevantním koncentracím chloridu kademnatého při sub-optimálním a optimálním přísunu sulfátů. Z výsledků diplomové práce vyplývá, že 50 µM CdSO4 více inhibuje růst kořenů klíčních rostlin Arabidopsis thaliana než 50 µM CdCl2, u tabáku tento efekt nebyl pozorován. Zvýšení hladiny transkriptů případně dřívější nástup transkripce genů kódujících proteiny účastnících se katabolických procesů v důsledku expozice kademnatým iontům nebyl u klíčních rostlin Arabidopsis thaliana a Nicotiana tabacum a hydroponicky pěstovaných rostlin Arabidopsis thaliana pozorován. U hydroponicky pěstovaných rostlin Nicotiana tabacum docházelo převážně ve starých listech k navýšení hladiny transkriptů námi analyzovaných genů, avšak tyto změny neodpovídaly fenotypovému projevu. Získané výsledky ukazují na jisté odlišnosti v reakci modelových rostlin na přítomnost kademnatých solí a jako součást širší studie by měly vést ke zpřesnění a rozšíření znalostí o odpovědi rostlin na abiotický stres.

Abstract:

In this diploma thesis, we deal with plant response to the abiotic stress caused by the cadmium exposure. This toxic metal has an adverse effect on plant growth and development, and therefore on the general agriculturally-relevant plant production. Cadmium-induced stress affects the basic plant metabolic pathways including the premature onset of aging. Leaf senescence is a regulated sequence of catabolic processes that lead to the cell death. Leaf aging is associated with extensive changes in gene expression. The aim of this work was to compare the effect of two different cadmium salts on the growth of roots of Arabidopsis thaliana and Nicotiana tabacum seedlings and to analyze transcript levels of selected genes involved in senescence processes (chlorophyll degradation – ABF4 and NYE1 in Arabidopsis thaliana; NYC1 and PAO in Nicotiana tabacum, and protein degradation – UBC1 and RPT1A in Arabidopsis thaliana, RPT1A in Nicotiana tabacum). Further, consequences of the long term exposure to the two environmentally relevant concentrations of cadmium salt under sub-optimal and optimal sulphate supply were studied using hydroponically cultivated Arabidopsis thaliana and Nicotiana tabacum plants. Our results show that presence of 50 µM CdSO4 in cultivation media inhibits the growth of Arabidopsis thaliana seedlings roots more severely compared to 50 µM CdCl2 while this effect was not observed during Nicotiana tabacum early development. Increased levels of transcripts and/or earlier activation of transcription of genes encoding proteins involved in catabolic processes indicating earlier onset of leaf senescence due to exposure to cadmium ions was not observed in Arabidopsis thaliana and Nicotiana tabacum seedlings and in hydroponically cultivated Arabidopsis thaliana plants. In hydroponically cultivated Nicotiana tabacum plants, the levels of transcripts of selected genes increased mainly in old leaves, but these changes did not correspond to the phenotypic changes. These results demonstrate distinct differences in responses of model plants to the exposure to cadmium salts and as a part of the complex study are supposed to refine our knowledge about the responses of plants to the abiotic stress.
