Cell migration in the biology and diagnostics of melanoma

Abstract:

V této bakalářské práci shrnuji současné poznatky o vývoji melanomu, migraci nádoru a tvorbě metastáz s ohledem na diagnostiku a terapeutické aplikace v budoucnu. Změny v signálních drahách a genetické aberace v melanocytech vedou k jejich maligní transformaci, nekontrolovatelné proliferaci, reakci imunitního systému a zvýšené migrační a invazivní schopnosti. Tyto vlastnosti zapříčiňují to, že je melanom nejagresivnějším nádorem kůže. Lepší pochopení jeho biologických vlastností a interakcí s mikroprostředím je klíčem pro úspěšnější léčebné strategií v budoucnu, tj. cílená, adjuvantní a kombinovaná terapie. Některé z těchto přístupů by však mohly vést k selekci rakovinných buněk, které budou na léčbu rezistentní, což by mělo za následek změnu chování a vlastností nádoru. Toto je také jedna z nejdůležitějších výzev, kterou by se měly zabývat biomedicínské studie. Celkově vzato, poslední roky ukázaly velký pokrok v léčbě melanomu, ale stále je zde prostor pro zlepšování a i v budoucnu je třeba zkoumat mechanismy nádorové biologie.

Abstract:

In this thesis I summarize the current knowledge about melanoma development, cancer migration and metastasis in perspectives for the future diagnostic and therapeutic applications. Changes in the signalling pathways and occurrence of genetic aberrations in melanocytes lead to their malignant transformation, uncontrolled proliferation, immunoreaction and increased migratory and invasive abilities. These features make melanoma the most aggressive skin cancer. Better understanding of its biology and interactions with microenvironment is a key for the perspective application of more successful treatment strategies, i.e. targeted, adjuvant therapies or combined treatments. Nevertheless, some of these approaches might lead to the selection of treatment resistant cancer cells, resulting in the change of cancer behaviour and properties. This is also one of the most important challenges for biomedical studies. Taken together, recent years showed a great progress in melanoma treatments, but still there is a space for improvement and various mechanisms of cancer biology need to be further investigated.
