Genetic characterization of honey bee populations

Abstract:

Abstrakt: Tato diplomová práce je zaměřena zejména na genetickou charakterizaci populace včely medonosné (Apis mellifera) na území České Republiky za použití metod molekulární biologie a genomiky. Metodická část řeší hlavně laboratorní postupy a metody statistického zpracování dat nukleotidových sekvencí genů cytochrom oxidáza I a cytochrom b. V poslední části vyhodnocuji výsledky mé práce v laboratoři, vytvořené evoluční stromy, hodnoty nukleotidové variace v rámci populace i mezi jejími skupinami založenými na geografickém původu vzorků a Hodnoty Tajimova testu neutrality. Výsledky mé práce zahrnují, sestavení fylogenetických stromů na základě nukleotidových sekvencí genů a segregačních bodů genů Cytochrom oxidáza I a Cytochrom b. Stanovení hodnoty variace populace v rámci nukleotidových sekvencí genů (πCOI = 0,002189), (πCyt-b = 0,011759). Stanovení hodnoty „Tajima´s D“ COI (-0,930616), Cyt-b (-0,644359). Určení hodnoty průměrné variace mezi skupinami podle geografické příslušnosti, cyt-b nejvyšší diverzita, (0,0132). Naopak nejnižší diverzita (0,0032), COI nejvyšší hodnoty, (0,00275) nejmenší, (0,00084).

Abstract:

Abstract: This diploma thesis is mainly focused on the genetic characterization of the honey bee population (Apis mellifera) in the Czech Republic using the methods of molecular biology and genomics. The methodical part deals mainly with laboratory procedures and methods of statistical data processing of nucleotide sequence data of cytochrome oxidase I and cytochrome b genes. In the last part I evaluate the results of my work in the laboratory, created evolutionary trees, nucleotide variation within the population, and between its groups based on the geographical origin of samples. and Tajima´s Neutrality Test Values. The results of my work include the construction of phylogenetic trees based on the nucleotide sequences of genes and segregation points of Cytochrome oxidase I and cytochrome b genes. Determination of the value of population variation within the nucleotide sequences of genes (πCOI = 0.002189), (πCyt-b = 0.011759).Determination of the value of "Tajima´s D" COI (-0.930616), Cyt-b (-0.644359). Determination of the value of the average variation between groups according to the geographical affiliation, cyt-b highest diversity (0.0132). On the contrary, the lowest diversity, (0.0032), at COI the highest value (0.00275) the lowest (0.00084).
