Sledování odolnosti rostlin vůči abiotickému stresu u rostlin s modulovanou expresí významných transkripčních faktorů

Tolerance to abiotic stress in plants with modulated expression of important transcription factors

Abstract:

V současné době je problém globální změny klimatu velmi aktuální. V důsledku zvyšujících se teplot musí rostliny čelit suchu a z něj plynoucímu osmotickému stresu v čím dál větší míře. Narušení přirozených podmínek má negativní dopad na růst a vývoj rostlin. Vzhledem k negativním dopadům těchto změn na produkci zemědělských plodin, je důležité lépe porozumět obraným mechanismům rostlin. V práci byla hodnocena odolnost vůči osmotickému stresu u linií vybraných z rostlinné kolekce Transplanta. Soubor zahrnoval linie s farmakologicky regulovatelnou expresí transkripčních faktorů WRKY, VOZ2, RAP2.12, NAC029 a RAV2. Analýza exprese regulovaných transkripčních faktorů pomocí RT-qPCR ukázala, že mezi jednotlivými liniemi jsou při působení 10µM β-estradiolu výrazné rozdíly v míře navýšení hladiny transkriptů cílových genů a u jedné linie bylo zjištěno dokonce snížení exprese při jeho aplikaci. Kultivace klíčních rostlin na médiu s β-estradiolem vedla k mírné inhibici růstu kontrolních rostlin v kontrolních i stresových podmínkách. Analýza morfologie prokázala, že transgenní linie s modulovatelnou expresí genu RAP2.12 mají podpořen růst nadzemní části klíčních rostlin i kořenů oproti standardním rostlinám, a to v podmínkách s i bez působení osmotického stresu. Tento výsledek však nebyl závislý na aplikaci β-estradiolu.

Abstract:

The problem of global climate change is very actual. As a result of rising temperatures, plants have to face increasing drought and the resulting osmotic stress. Abiotic stress harms plant growth, development and plant production. Thus, it is important to understand plant defence mechanisms to limit crop losses. Here, the tolerance to osmotic stress of selected transgenic plants from Transplanta collection was studied. The set included lines with a pharmacologically controllable expression of transcription factors WRKY, VOZ2, RAP2.12, NAC029 and RAV2. Analysis of the expression of transcription factors by RT-qPCR showed that there were significant differences between the lines of 10µM β-estradiol in the rate of increase in the level of transcripts of target genes, and one line even showed a decrease in expression when applied. Cultivation of seedlings on medium with β-estradiol led to a slight inhibition of the growth of control plants in control and stress conditions. Morphology analysis showed that transgenic lines with a regulated expression of the RAP2.12 gene have supported the growth of the aboveground part and roots of seedlings compared to wild type plants, in conditions with and without osmotic stress. However, this result was not dependent on β-estradiol application.
