Nanotransporters for targeted transport of cytostatics across the blood-brain barrier

Abstract:

Tato diplomová práce je zaměřena na studium nanotransportérů vhodných pro cílenou dopravu přes hematoencefalickou bariéru při nádorové terapii centrální nervové soustavy. Literární část pojednává o tématu využití nanočástic v terapii nádorů a možnostech jejich přenosu přes hematoencefalickou bariéru. Hlavním tématem jsou nádorová onemocnění centrální nervové soustavy a možnosti jejich léčby. V současnosti jsou nanotransportéry předmětem výzkumů jako možnost šetrné léčby nádorového onemocnění. V této práci byly zkoumány cytotoxické účinky enkapsulovaného doxorubicinu na nádorové buňky. Biologicky aktivní molekuly doxorubicinu byly enkapsulovány do feritinu. Díky enkapsulaci došlo k posílení léčivého účinku doxorubicinu a zároveň byly ochráněny nenádorové buňky. Enkapsulační výtěžnost byla 20–40 %. U výsledných nanokonstruktů byly charakterizovány spektrální vlastnosti, velikost, ζ-potenciál. Pro stanovení optimálního sestavení nanokonstruktu byla provedena nativní PAGE. Výsledky byly použity pro vyhodnocení vhodnosti využití výsledných nanokonstruktů pro nádorovou terapii. Dle výsledků se jako optimální jeví feritin PFU z archea Pyrococcus furiosus, u kterého byla zjištěna optimální velikost 13 nm a nejvyšší enkapsulační výtěžnost, a to 38,47 %. Jako optimální se jeví také feritin ECaLH izolovaný ze sleziny Equus caballus, u kterého byla zjištěna velikost 13 nm a enkapsulační výtěžnost 30,99 %. Pomocí buněčných testů byla testována cytotoxicita výsledných nanokonstruktů vůči buněčným liniím HBL-100, MDA-MB-231, MCF-7, T-47D, U-87, ZR-75-1. Vysokou míru cytotoxicity vykazoval nanokonstrukt ECaLH+doxorubicin vůči nádorové buněčné linii glioblastomu U-87. Po aplikaci toho-to nanokonstruktu byla průměrná viabilita buněk 34 %, tedy ještě nižší než v případě volného doxorubicinu. To značí, že doxorubicin enkapsulovaný v tomto nanokonstruktu by mohl být slibným léčivem v cílené nádorové terapii centrální nervové soustavy.

Abstract:

The aim of this thesis is to study nanocarriers suitable for directed transport through blood-brain barrier in tumour therapy of central nervous system. The literary part is focused on the use of nanoparticles in anti-tumour therapy and their possible transport through blood-brain barrier. The main topic is tumour diseases of central nervous system and their treatment options. Nowadays, nanocarriers are the subject of research as a new method of gentle treatment of tumour diseases. In this work the cy-totoxic effects of encapsulated doxorubicin on tumour cells were examined. Biologi-cally active molecules of doxorubicin were encapsulated into ferritin. The encapsula-tion aimed at improving the therapeutic effect of doxorubicin and while simultaneous-ly protecting the non-tumour cells. Encapsulation yield was 20–40 %. For resulting nanoconstructs, spectral properties, size, and ζ-potential were defined. The native PAGE was performed to determine optimal assembly of the nanoconstruct. The results were used for evaluating the suitability of the final nanoconstructs in anti-tumour ther-apy. According to the results, ferritin PFU from archaea Pyrococcus furiosus seems to be the optimal choice, for which an optimum size of 13 nm, and highest encapsulation yield of 38,47 % were established. Another optimal choice seems to be ferritin ECaLH isolated from spleen Equus caballus, for which a size of 13 nm and encapsulation yield of 30,99 % were established. Using in vitro tests the cytotoxicity of final nanocon-structs towards cell lines HBL-100, MDA-MB-231, MCF-7, T-47D, U-87, ZR-75-1 was tested. Nanoconstruct ECaLH+doxorubicin was showing a high degree of cytotox-icity towards the cell line U-87. After applying this nanocomplex, the average cell via-bility was 34 %, even higher than after treatment with free doxorubicin. This proves that doxorubicin encapsulated in this nanoconstruct could be a promising medicament in anti-tumour therapy of central nervous system.
