Use of special staining and immunohistochemical methods for the detection of Helicobacter pylori

Abstract:

Bakalářská práce je zaměřena na využití speciálních barvících a imunohistochemických metod v průkazu Helicobacter pylori. HP je pohyblivá, gramnegativní, spirální bakterie, která kolonizuje žaludeční sliznici. Cílem práce bylo přehledně zpracovat používané metody v průkazu HP v histologické laboratoři. Dalším cílem bylo srovnat tyto metody z hlediska náročnosti provedení, vybavenosti laboratoře, času a finanční stránky. V teoretické části práce popisuji charakteristiku HP, patogenezi, epidemiologii infekce HP, přehled onemocnění vyvolané HP, léčbu a metody průkazu. Objasněna je zde také cesta bioptického vzorku do histologické laboratoře, jeho zpracování a také samotné metody průkazu s vysvětlením jejich principu. V rámci praktické části jsou zahrnuty přesné metodické postupy, výsledky barvení a fotodokumentace. Ze speciálních barvících metod využívám barvení Giemsa-thionin, impregnaci dle Warthin-Starry, toluidinovou modř. Rovněž popisuji imunohistochemickou metodu s využitím polyklonální protilátky anti-Helicobacter pylori. Praktická část byla provedená na Ústavu patologie Fakultní nemocnice v Ostravě.

Abstract:

Bachelor thesis is focused on the use of special staining and immunohistochemical methods for detection of Helicobacter pylori. HP is a moving, gram-negative, spiral bacterium that colonizes the gastric mucosa. The aim of the thesis was to elaborate the methods used for detection of the HP in the histological laboratory. Another aim was to compare these methods in terms of execution difficulty, laboratory facilities, time and financial aspects. In the theoretical part of the thesis I describe HP characteristics, pathogenesis, HP infection epidemiology, overview of HP-induced diseases, treatment and methods of identification. I also describe the journey of the biopsy specimen to the histological laboratory, its processing, and also the methods of the diagnosis with an explanation of their principle. The practical part includes precise methodological procedures, staining results and photo documentation. I use Giems-thionin staining, Warthin-Starry impregnation, toluidine blue staining. I also describe an immunohistochemical method using polyclonal antibody against Helicobacter pylori. The practical part was done at the Institute of Pathology of the Faculty Hospital in Ostrava.
