Cultivation of cryptosporidia in cell and cell-free cultures

Abstract:

Zástupci rodu Cryptosporidium (Apicomplexa) jsou jednou z hlavních příčin těžkých průjmů a dětské úmrtnosti v rozvojových zemích. Bohužel proti tomuto původci nemoci zvané kryptosporidióza, převážně invadující gastrointestinální trakt, nejsou v současné době k dispozici žádná dlouhodobě účinná léčiva. Předkládaná diplomová práce se zabývá velmi aktuálním tématem kultivace zástupců rodu Cryptosporidium v podmínkách in vitro. Vyvinutí funkčního in vitro systému umožňujícího dlouhodobou kultivaci těchto parazitů by umožnilo prohloubit znalosti o jejich patofyziologii a tím by se také významně zvýšila šance na vyvinutí účinných antikryptosporidiálních léčiv. Teoretická část diplomové práce stručně popisuje rod Cryptosporidium a jeho životní cyklus. Tato část také shrnuje různorodé faktory, které mají zásadní vliv na úspěšnou kultivaci kryptosporidií v in vitro podmínkách. Zmíněná teoretická část se nevěnuje pouze kultivaci na buněčných kulturách, ale také možnosti kultivace v čistém médiu za absence hostitelských buněk (tzv. bezbuněčné neboli axenické kultury). Praktická část práce navazuje na bakalářskou práci a věnuje se kultivaci kryptosporidií na buněčných kulturách. Další část práce se zabývá testováním možností kultivace s využitím bezbuněčných kultur na základě publikovaných protokolů. Při laboratorní práci byl využit střevní druh Cryptosporidium parvum a žaludeční patogen Cryptosporidium proliferans.

Abstract:

Members of the genus Cryptosporidium (Apicomplexa) are the leading cause of diarrhoea and a major cause of child mortality worldwide in developing countries. This agent causes a disease called cryptosporidiosis and predominantly invades the gastrointestinal tract. Unfortunately, no long-effect drugs are currently available against cryptosporidian. The following master's thesis deals with a very current topic of in vitro cultivation of Cryptosporidium spp. Developing a functional in vitro system allowing the long-term cultivation of these parasites would enable to deepen the knowledge of their pathophysiology and this way significantly increase the chances of developing effective anti-cryptosporidial drugs. The theoretical part of the master's thesis briefly describes the genus Cryptosporidium and its life cycle. This section also summarises various factors that have a major impact on the successful cultivation of cryptosporidia under in vitro conditions. This theoretical part does not only deals with parasite cultivation in cell cultures but also with possibilities of its cultivation in a pure medium with the absence of host cells (the so-called cell-free or axenic cultures). The practical part of the thesis follows the bachelor thesis and goes into details about the cultivation of cryptosporidia in cell cultures. Next part of the thesis deals with the testing of cultivation possibilities using the cell-free cultures based on so far published protocols. During the laboratory work, the intestinal species Cryptosporidium parvum and the gastric pathogen Cryptosporidium proliferans were used.
