Interaction of chelerythrine with d(GGAAGCTTCC)2

Abstract:

Teoretická část práce shrnuje obecné poznatky o chelerythrinu a kvartérních benzo[c]fenanthridinových alkaloidech, poukazuje na jejich významné biologické účinky a možnosti využití těchto alkaloidů v klinické praxi. Jako jedno z významných působení CHE na buněčné struktury je diskutována interakce s DNA. Pomocí metod molekulového modelování byly zobrazeny tři různé způsoby interakce CHE s DNA; interkalace a pro porovnání vazba do malého žlábku se dvěma různými orientacemi molekuly CHE. Na základě molekulárně mechanických výpočtu byly určeny vazebné energie jednotlivých komplexací. Na konec byla provedena MD simulace volného oligonukleotidu a výpočet energie potřebné ke změně konformace při procesu interkalace.

Abstract:

The theoretical part of thesis summarizes the general knowledge of the chelerythrine and quaternary benzo[c]phenanthridine alkaloids, points to their significant biological effects, and aplication of these alkaloids in clinical practice. Interaction with DNA as one of the dominant effects of CHE on cell structure is discussuted. Using molecular modeling three different ways of interaction of CHE with DNA were shown. These are intercalation and minor groove binding with two different molecular orientations of CHE. On the basis of molecular mechanical calculation, binding energy of each complexation was determined. In addition, the energy of conformational change of free oligonucleotide was calculated.
