Characterisation of Pseudomonas aeruginosa strains isolated from infections of urinary tract

Abstract:

Nozokomiální infekce představují významný problém při léčbě imunokompromitovaných pacientů, a to nejen z hlediska komplikace samotné léčby, ale také z ekonomického hlediska. Močové infekce způsobené bakterií Pseudomonas aeruginosa jsou druhým nejčastějším onemocněním spojeným s hospitalizací pacienta. Jako alternativa k léčbě antibiotiky jsou v současnosti zvažovány pyociny, antibakteriální proteiny produkované pseudomonádami. V předložené práci je popsáno testování souboru 140 klinických izolátů pocházejících z infekcí močového traktu na produkci pyocinů. K jejich charakterizaci byla použita fenotypová a genotypová metoda. Fenotpová metoda prokázala vysokou míru antagonismu mezi kmeny navzájem. Pomocí PCR testování bylo zjištěno, že až 96,4 % klinických izolátů nese geny pro alespoň jeden z jedenácti testovaných pyocinů. Srovnáním genotypové a fenotypové metody bylo potvrzeno, že pyociny mají velký potenciál být využity jako alternativní terapeutické látky při léčbě pseudomonádových infekcí.

Abstract:

Nosocomial infections represent a significant problem in a treatment of immunocompromised patients. Not only in the term of treatment itself but also in the economic point of view. Urinary infections caused by Pseudomonas aeruginosa are the second most common diseases associated with the patient´s hospitalization. These infections most commonly occur as a secondary infections after insertion of urinary catheter. In most cases cathetrisation is not necessary at all. P. aeruginosa are equipped with various virulence determinants that can increase the resistence of bacteria to the commonly used therapeutics. Pyocins are a suitable alternative to the antibiotics. They belong in a group of bacteriocins produced by pseudomonads with a narrow killing range. The aim of this work was to investigate the pyocinogeny in the 140 P. aeruginosa clinical isolates from catheter infections. A PCR detection was used for molecular detection of pyocins after a phenotypic screening that showed a great degree of antagonism between the strains. Using PCR testing, it was found that 96,4 % of clinical isolates carry genes for at least one pyocin out of eleven. By comparing the genotypic and phenotypic methods, it was confirmed that pyocins have great potential to be used as alternative therapeutic agents in the treatment of pseudomonas infections.
