Methods for identification of \kur{"Arcobacter-like"} microorganisms

Abstract:

Diplomová práce se zabývá patogenními bakteriemi Arcobacter spp. a jejich identifikací. Teoretická část shrnuje informace o tomto bakteriálním rodu, o jeho charakteristických vlastnostech, výskytu, patogenitě a faktorech virulence. Dále jsou zde obsaženy kapitoly o kultivačních metodách a molekulárně-biologických metodách identifikace. Cílem experimentální části práce bylo identifikovat sbírkové kmeny těchto bakterií na základě různých PCR protokolů s různou identifikační schopností. Dále byla optimalizována 16S rRNA-RFLP metoda pro identifikaci zástupců čeledi Arcobacteraceae. Všechny identifikační postupy byly aplikovány také na kmeny arkobakterů izolované z vzorků izolovaných z potravin a zdrojů vod. Na základě získaných výsledků byly jednotlivé metody porovnány a jejich vhodnost k identifikaci arkobakterů diskutována.

Abstract:

The master thesis deals with pathogenic bacteria Arcobacter spp. and their identification. The theoretical part summarizes information about this bacterial genus, about its characteristics, occurrence, pathogenicity and virulence factors. There are also chapters about cultivation method and molecular-biological methods of identification. The aim of the experimental part of the study was to identify collection strains of these bacteria on the basis of different PCR protocols with different identification capabilities. Furthermore was optimized the 16S rRNA-RFLP method for the identification the representatives of Arcobacteracea. All identification procedures were also applied to arcobacter strains isolated from samples isolated from food and water sources. Based on the obtained results, the individual methods were compared and their suitability for the identification of arcobacteria was discussed.
