Study of Antioxidant Components in Grape Seed Extracts

Abstract:

V této práci byl optimalizován proces extrakce hroznových semen (Vitis vinifera L.). Tato optimalizace probíhala za využití odrůdy Cerason. Byly sledovány tyto parametry: poměr matrice vůči ethanolu 1:05, 1:10 a 1:15, koncentrace ethanolu 25, 50 a 75 % obj. a délka macerace 72, 168 a 240 hodin. Bylo tedy vytvořeno 27 variant, u kterých byla po uplynutí macerace stanovena antioxidační aktivita metodami ABTS, DPPH, FRAP, Free Radicals a obsah celkových polyfenolických sloučenin. Nejlepší varianta se ukázala s kombinací 1:05, 75 % obj. ethanol a 240 hodin. Dále byly, pomocí HPLC, stanoveny vybrané polyfenolické sloučeniny. Největší koncentrace byly zjištěny u epicatechinu, kyseliny gallové a catechinu. Kromě jiného byly také porovnávány hodnoty antioxidačního potenciálu u odrůd Laurot, Merlot, Carmenére, Tramín červený a Hibernal. Hroznová semena byla podrobena extrakci podle nejlepšího výsledku z optimalizace procesu extrakce (poměr 1:05, 75 % obj. ethanol, délka macerace 240 hodin). Nejvyšších hodnot obsahu antioxidantů dosáhly odrůdy Tramín červený a Laurot. Naopak odrůda Hibernal měla nejnižší hodnoty. Dále se srovnávaly obsahy antioxidantů u odrůdy Cerason mezi ročníky 2016 a 2017. Byla stanovena antioxidační aktivita a obsah celkových polyfenolických sloučenin. Ve všech případech byl jednoznačně nejlepší rok 2016. Na závěr se sledovala stabilita lyofilizovaných extraktů odrůdy Cerason z optimalizace procesu extrakce. Rozhodujícími parametry byla teplota (20 a 30 °C) a čas (0, 3, 6, 9, 12 měsíců). V průběhu času dochází k mírnému poklesu hodnot, který ale není nijak velký. Průměrně hodnoty antioxidační aktivity klesaly v rozmezí o 1,18–2,10 g/l a obsah celkových polyfenolických sloučenin v rozmezí o 0,13–0,88 g/l.

Abstract:

The process of grape seed extraction (Vitis vinifera L.) has been optimized in this work. This optimization was carried out using the Cerason variety. The following parameters were monitored: matrix to ethanol ratios of 1:05, 1:10 and 1:15, ethanol concentration 25, 50 and 75% vol., and maceration lengths 72, 168 and 240 hours. Thus, 27 variants were developed for which the antioxidant activity was determined by ABTS, DPPH, FRAP, Free Radicals and total polyphenolic compounds after maceration. The best variant showed a combination 1:05, 75 % vol. ethanol, and 240 hours. Furthermore, selected polyphenolic compounds were determined by HPLC. The highest concentrations were measured for epicatechin, gallic acid and catechin. Besides, the antioxidant potential values of Laurot, Merlot, Carmenére, Tramín červený and Hibernal were also compared. Grape seed of these varieties was used: Laurot, Merlot, Carmenerer, Tramin Red and Hibernal. The grape seeds were extracted by using this procedure: ratio 1:5, ethanol 75 % vol., maceration length 240 hours, which was the best from optimization of the extraction process. Antioxidant activity was determined using ABTS, DPPH, FRAP, Free Radicals methods and total polyphenol compounds. The highest values of antioxidant content have been reached in Laurot and Tramín červený varieties. On the contrary, the Hibernal variety had the lowest values. Also, the antioxidant content of the Cerason variety was compared between the years 2016 and 2017. Antioxidant activity was determined and the content of total polyphenolic compounds. In all cases, the year 2016 was clearly the best. Finally, the stability of the lyophilized extracts of the Cerason variety from the optimization of the extraction process was monitored. The decisive parameters were temperature (20 and 30 °C) and time (0, 3, 6, 9, 12 months). Over time, there is a slight decrease in values, which is not very large. On average, the antioxidant activity values decreased in the range of 1.18–2.10 g/l and the total polyphenolic compounds decreased in the range of 0.13–0.88 g/l.
