Studying the Effect of Heavy Metals on Cell Respiration

Abstract:

Diplomová práce se zabývá vlivem toxicity kadmia na buněčnou respiraci in vitro. V teoretické části je stručně popsána mitochondrie, mitochondriální komplexy a jejich inhibitory. Také je zde popsána funkce rozpojovacích proteinů. Následně jsou v kapitolách popsány možnosti měření buněčné respirace, druhy biologických materiálů a přehled buněčných linií, které byly v posledních letech nejčastěji využívány ve vysokoúčinné respirometrii. V druhé polovině teoretické části je obsah zaměřen na těžké kovy, specificky na kadmium a na jeho působení na úrovni organismu, buňky a mitochondrií. Experimentální část práce se zabývá přípravou biologických materiálů pro měření respirace. Mezi použitými biologickými materiály jsou PBMC, buněčné linie Jurkat, A549, HK-2 a izolované mitochondrie. Pro použití PBMC bylo nejprve potřeba zavést metodu izolace buněk z periferní krve. A optimalizovat parametry metody tak, aby bylo získáno maximální množství PBMC v co největší čistotě. Pro studium vlivu CdCl2 na respiraci bylo také potřeba optimalizovat koncentraci digitoninu potřebnou pro permeabilizaci cytoplazmatické membrány jednotlivých druhů buněk. Permeabilizované buňky a izolované mitochondrie byly poté využity k testování respirace po přímém přídavku CdCl2 do komor respirometru. Intaktní buňky byly inkubovány s 100 mol/l CdCl2 po dobu 2 a 4 hodin. Z výsledků získaných při měření na respirometru Oroboros vyplývá, že toxicita CdCl2 stoupá s dobou inkubace buněk s CdCl2.

Abstract:

The thesis deals with the effects of cadmium toxicity on cellular respiration in vitro. The theoretical part briefly describes mitochondria, mitochondrial complexes and their inhibitors. It also describes the function of uncoupling proteins. Other chapters have been focused on the possibilities of cell respiration measurement and the types of biological materials, including an overview of cell lines that have been used in high-resolution respirometry most frequently in recent years. Last part of the Introduction focuses on the heavy metals, especially on cadmium and its effects in organism, cells and mitochondria. The experimental part of the thesis deals with preparation of biological materials for measurement of cell respiration by high-resolution respirometry. We used a variety of biological material, e.g. PBMCs, Jurkat cells, A549, HK-2 and isolated mitochondria. In order to use PBMC, the method of cell isolation from peripheral blood had to be introduced and then optimized in order to obtain the maximal content of PBMCs of the highest purity. In order to study the influence of CdCl2 on respiration, it was also necessary to optimize the concentration of digitonin required for the permeabilization of the cytoplasmic membrane of individual cell types. Afterwards, permeabilized cells and isolated mitochondria were used to evaluate respiration after acute addition of CdCl2 to the respirometer chambers. Intact cells were incubated with 100 mol/l CdCl2 for 2 and 4 hours. The results obtained from the Oroboros respirometer show that CdCl2 toxicity is increasing with time of cell incubation with CdCl2.
