Utilisation of imaging of elements in bioapplications

Abstract:

Předkládaná dizertační práce se věnuje zobrazování prvků/kovů v biologických tkáních pomocí techniky laserové ablace s indukčně vázaným plazmatem a hmotnostní spektrometrií (LA-ICP-MS). V posledních letech zájem o zobrazení distribuce těchto kovů vzrostl, a to zejména v biomedicínské oblasti, dnes často také zmiňované „life science“ (věda o živé přírodě), nebo přímo v metalomice (vědecká oblast zahrnující komplexní analýzu celého spektra elementárních druhů uvnitř buňky, či tkáně). První část klade důraz na stanovení distribuce esenciálních prvků (např. Cu, Zn, Fe), těžkých kovů (Pb – spojených s bioakumulací, Pt – jako součást chemoterapeutik a její působení) pomocí LA-ICP-MS, jejich kvalitativního i kvantitativního stanovení v biologických vzorcích (nádory kůže a prostaty, či tkáně tasemnice). V současné době navíc roste zájem o zobrazování nejen prvků, ale také proteinů, ve kterých jsou tyto prvky vázány a také jimi transportovány. Proto se druhá část této práce věnuje vyvinutí nové metody pro stanovení proteinů pomocí LA-ICP-MS. Konktrétně se jedná o kombinaci metod immunohistochemie (značení protilátek kovovou značkou – nanočásticí např. Au, Ag, či lanthanoidy, která lze pomocí LA-ICP-MS zobrazit, a následné selektivní navázání protilátky na zájmový protein), s LA-ICP-MS umožňující proteiny zobrazit. Jako modelový protein byl zvolen myší protein IgG. Po ověření funkčnosti této vyvinuté metody je dále uvedena její aplikace na reálný vzorek se stanovením proteinu p53 - nádorový marker. Tato metoda ovšem nemusí být vhodná pro všechny typy proteinů, jako v případě malých proteinů typu metallothionein (MT, 6-10 kDa), a to z důvodu jejich nízké hmotnosti, či nedostupnosti dané protilátky. Proto je zde představena další nová kombinace metod LA-ICP-MS s využitím tzv. molekulově imprintovaných polymerů (MIP), díky kterým je možno MT stanovit a navíc od sebe odlišit jejich jednotlivé typy (MT1 a MT3). Jelikož stanovení distribuce prvků v biologických tkáních spolu s jejich standardizací v posledních letech velice pokročilo, závěrečná část této práce se věnuje přípravě laboratorní úlohy pro magisterské studenty - Zobrazení distribuce a kvantifikace esenciálních prvků (Cu, Zn) a platiny (z citostatického léčiva) v tenkých řezech nádoru prostaty.

Abstract:

This thesis summarizes results obtained during my doctoral study. It is focused on imaging of elements/metals in biological tissues by means of laser ablation with inductively coupled plasma and mass spectrometry (LA-ICP-MS). In the last decades increased the interest about imaging of these metals distribution, mainly in biomedical research, life science or specifically in metallomics (research concerning the complexing analysis of the whole spectrum of elementary species inside the cell or tissue). The first part is devoted to the determination of distribution of essential elements (Cu, Zn, Fe), heavy metals (Pb – connecting with bioaccummulation, Pt – as a part of cancer drugs and their effects) by means of LA-ICP-MS, their qualitative and quantitative determination in biological samples (tumor and prostate tissues, or tapeworm tissues). Moreover, there is a growing interest in bioimaging not only elements but also proteins. These proteins bind interested elements, and even they transported them. Therefore, the second part of this thesis is focused on the development of a new strategy of protein determination by means LA-ICP-MS. Particularly speaking about a combination of two method, immunochemistry (labelling antibodies by metal code – nanoparticles such as Au, Ag, or lanthanides, which can be imaged/determine by LAICP-MS, and subsequently selective binding of an antibody to interested protein), with LA-ICP-MS enabling to image proteins. As model protein was chosen the mouse IgG protein. After verifying the functionality of this developed method, there is presented the application to real sample with p53 protein determination – as tumor marker. However, this method may not be suitable for all types of proteins, such as small proteins metallothioneins (MT, 6-10 kDa) because of their low mass or the antibody unavailability. Hence, next is presented other new combination of methods, LA-ICPMS with the use of molecularly imprinted polymers (MIP), which make it possible to determine MT and moreover to distinguish their two individual types (MT1 and MT3). Since the distribution of elements in biological tissues together with their standardization moved forward in last years, the last part of this thesis is dedicated to preparation of lab task for pregraduate students – Imaging of essential elements (Cu, Zn) and platinum (from cancer drug) distribution and their quantification in thin sections tissues of prostate tumor.
