Isolation and characterization of mutant S. aureus strains sensitive to the polyvalent phage 812

Abstract:

Bakteriální druh Staphylococcus aureus je běžnou součástí mikroflóry člověka, avšak v důsledku oslabení imunitního systému hostitele může být původcem závažných infekcí. V souvislosti s globálním rozšířením kmenů S. aureus rezistentních k dostupným antibiotikům roste zájem o využití bakteriofágů při boji s tímto patogenem. Pro tyto účely jsou vhodné polyvalentní virulentní bakteriofágy, které jsou schopné napadat většinu kmenů S. aureus. Kmeny necitlivé vůči terapeutickým fágům však mohou způsobit selhání léčby. Objasnění mechanismů navozujících rezistenci kmenů S. aureus k fágům může umožnit přípravu terapeutických fágů s rozšířeným rozmezím hostitele. Diplomová práce je zaměřena na studium mechanismů podmiňujících necitlivost kmenů S. aureus vůči polyvalentnímu fágu 812. Z kmenů S. aureus rezistentních k fágu 812 byly chemickou mutagenezí připraveny mutanty s navozenou citlivostí vůči tomuto fágu. Získané mutanty byly dále charakterizovány a jejich vlastnosti porovnány s vlastnostmi příslušných standardních kmenů. Uvedeným přístupem se podařilo prokázat, že rezistence studovaných kmenů S. aureus vůči infekci fágem 812 není navozena blokací adsorpce fága k hostitelské buňce ani přítomností systémů CRISPR/Cas. Jako nejpravděpodobnější příčina necitlivosti se jeví činnost R M systémů, případně blokace injekce fágové DNA navozená přítomností profágů kódujících specifické proteiny.

Abstract:

The bacterial species Staphylococcus aureus can be found as a normal part of human microflora, but at times when the immunity system of the host is reduced, it can be the agent of serious infections. Due to global distribution of the S. aureus strains resistant to available antibiotics, there is an increasing interest in using bacteriophages to fight this pathogen. Polyvalent virulent bacteriophages seem suitable for these purposes, because they are capable of attacking most of the S. aureus strains. Strains insensitive to therapeutic phages, however, can lead to the failure of the treatment. Elucidation of the mechanisms responsible for the resistance of S. aureus strains against phages could allow preparing therapeutic phages with extended host range. This diploma thesis focuses on studying the underlying mechanisms of the insensitivity of S. aureus strains against the polyvalent phage 812. Using chemical mutagenesis, S. aureus strains resistant to phage 812 were used to prepare mutants with induced sensitivity for this phage. These mutants were subsequently further characterized and their properties compared with properties of their corresponding standard strains. Using this procedure it was possible to prove that the resistance of the studied S. aureus strains against phage 812 infections is not induced by blocking the phage adsorption to the host cell or by the presence of the CRISPR/Cas systems. The most likely cause of insensitivity seems to be the activity of R-M systems, alternatively the blocking of the injection of phage DNA due to the presence of prophages coding specific proteins.
