Therapeutic potential of intracellular signaling pathways modulation in malignant melanoma cells

Abstract:

Maligní melanom je zhoubný kožní nádor s vysokým metastatickým potenciálem. Incidence maligního melanomu v České republice vzrostla za posledních třicet let pětinásobně. V etiopatogenezi se uplatňují především genetické dispozice a vliv slunečního záření. Protože stávající možnosti terapie pokročilých stádií melanomu jsou velmi omezené a jejich prognóza je velmi špatná, je třeba pro tento typ nádoru nalézt nové, účinnější terapeutické postupy. Současný výzkum se zaměřuje na inhibici důležitých signálních drah, zejména RAS/RAF/MEK/ERK mitogenně aktivované signální dráhy. Tato dráha se zdá být klíčovou při rozvoji maligního melanomu a je často aktivována mutacemi v protoonkogenech NRAS nebo BRAF. Cílem této diplomové práce bylo studium možného terapeutického ovlivnění maligního melanomu pomocí nízkomolekulárních inhibitorů ovlivňujících vybrané regulační proteiny, zahrnující cyklin-dependentní kinázy (CDK), mitogenně aktivované signální dráhy ERK (MAPK) a nádorový supresor p53. Inhibitor CDK roskovitin je účinným inhibitorem CDK spřažených s aktivací buněčného cyklu. Kromě inhibice CDK je navíc schopen snižovat transkripci a senzitizovat některé nádorové linie k aktivaci apoptózy. Roskovitin je schopen také aktivovat tumor supresorový protein p53 a účinně spolupracovat s dalšími, v terapii již používanými cytostatiky. V našich experimentech jsme se rozhodli otestovat vliv současné aplikace roskovitinu v kombinaci s několika přírodními cytostatiky na melanomovou buněčnou linii. Výsledky naznačují, že dochází k výraznému snížení životaschopnosti melanomové linie při použití kombinace roskovitinu spolu s izoflavonoidem genisteinem a inhibitorem anti-apoptotických proteinů rodiny Bcl-2 gossypolem. Naopak kombinací CDK inhibitoru roskovitinu s mikrotubulárními toxiny docházelo ke snížení vlastní cytotoxicity cytostatik taxolu a vinblastinu na melanomovou buněčnou linii. Výsledky této práce tedy naznačují, že inhibitor CDK roskovitin by se mohl stát důležitým nástrojem pro boj s nádorovými onemocněními, je však nutné volit vhodně jeho kombinace s jinými protinádorovými léčivy.

Abstract:

Malignant melanoma is a malignant skin tumor with a high metastatic potential. The incidence of malignant melanoma in Czech republic has increased five times over the last 30 years. It is the genetic predisposition and the effect of solar radiation that are predominant in the etiopatogenesis. As the current treatment options for advanced melanoma remain unsatisfactory and their prognosis is very poor, there is clearly a need for the design of new, more efficient therapies for malignant melanoma. The current anti-melanoma research is mostly focused on the inhibition of crucial cellular pathways, especially the RAS/RAF/MEK/ERK mitogen-activated protein kinase pathway. This MAPK pathway has turned out to be critically important for the growth of melanoma since the proliferation of most melanomas is driven by somatic mutations of NRAS and BRAF proto-oncogenes. The aim of this diploma thesis was to test the possibility of the therapeutically influencing of the melanoma cell growth and survival using small molecule inhibitors of important cellular regulatory proteins, including cyclin-dependent kinases (CDKs), mitogen activated protein kinases (MAPKs) and tumor suppressor p53. CDK inhibitor roscovitine can directly interfere with cellular proliferation by inhibiting cell cycle progression. In addition to that, CDK inhibitor roscovitine has the ability to inhibit transcription and to sensitize some types of cancer to apoptosis. Roscovitine can also induce tumour suppresor p53 activation and can synergistically cooperate with other therapeutically used cytostatics drugs. In our experiments, we have tested the response of melanoma cell lines to the combination of roscovitine with several natural cytostatic agents. The results show that roscovitine cooperates with the dietary isoflavonoid genistein or inhibitor of anti-apoptotic proteins of the Bcl-2 family gossypol and promotes melanoma cell death. Another effective combination seems to be roscovitine with inhibitors of DNA topoisomerases where significantly reduces the viability of melanoma cells. Surprisingly, roscovitine had an opposite effect on the activity of microtubular toxins. The addition of roscovitine decreased the toxicity of taxol or vinblastine towards the melanoma cells. In conclusion, the results of this thesis suggest that CDK inhibitor roscovitine could be used as an efficient anti-melanoma agent but certain drug combinations should be avoided.
