Effects of compounds and their mixtures in aquatic environment on early development of fish

Abstract:

Environmentální polutanty antropogenního i přírodního původu představují dlouhodobý problém ve vodním prostředí a hodnocení a řešení rizik s nimi spojených závisí na toxikologických datech. Je tu stálý požadavek na další informace a data o nebezpečnosti pro zvyšující se množství látek, zatímco je zároveň tlak na snížení objemu testů toxicity na zvířatech. To vede ke zvýšené míře využívání alternativních metod, které nepoužívají dospělá zvířata jako in silico nebo in vitro metody. Tato disertace se věnuje využití embryonálního modelu Danio rerio, malé ryby z čeledi Cyprinidae, která má řadu vlastností, kterými je velmi vhodná pro testování toxicity. Obecným cílem této práce je vývoj a aplikace nových metod studia efektů environmentálních polutantů se zaměřením na problematiku vodního prostředí a ryb a mechanismů toxicity, které by mohly stát za negativními účinky, které by mohly nastat v prostředí. Pro tento účel byl použit embryonální test s Danio rerio pro charakterizaci toxicity antropogenních polutantů jako pesticidů a směsí jako odpadních vod nebo sinicových extraktů. Aplikovali jsme nové přístupy pro studium teratogenity extraktů z vodních květů sinic v kombinaci s detailní charakterizací a frakcionací směsí a s testováním receptorové aktivity in vitro (retinoidní receptor) s cílem identifikovat konkrétní látky přispívající k teratogenním účinkům (Studie I a II). V této práci jsme popsali, že frakce sinicových extraktů obsahující retinoidní látky jsou zodpovědné za drtivou většinu pozorované toxicity dané směsi. Na základě našich výsledků jsme mohli ukázat spojitost mezi retinoidními látkami v extraktech a teratogenitou, která byla pozorována u obratlovců. Také jsme detekovali neurotoxické efekty frakcí extraktů obsahující retinoidy, zde jsme však nebyli schopni tyto efekty spojit s konkrétními látkami a role retinoidů zde zůstává nejasná. Pro studium účinků široce používaných pesticidů jsme využili rozšířený embryo test s Danio rerio pro sledování vlivu na různé orgány a zahrnuli jsme i měření mechanistických parametrů jako kardiotoxicity pro objasnění účinků široce používaných pesticidů (Studie III a V). Také jsme naměřili zajímavá data ovlivnění molekulárních biomarkerů po expozici environmentálním koncentracím pesticidů. Ukázali jsme, že embryonální test s Danio rerio představuje velmi dobrý nástroj pro hodnocení rizik chemických látek vzhledem k tomu, že může poskytnout jak informace o klasických parametrech jako jsou deformace nebo mortalita, tak o mechanistických parametrech, po kterých je stále se zvyšující poptávka obzvlášť ze strany evropských regulátorů. Také jsme embryo test s rozšířením sledovaných parametrů využili pro charakterizaci komplexních směsí – odpadních vod z těžby ropy („produced water“) doplněných zevrubnou chemickou analýzou (Studie IV). Ukázali jsme, že testování pomocí biologického modelu funguje jako užitečný doplněk chemické analýzy u komplexních směsí vzhledem k tomu, že změření všech složek a odhad toxicity na základě složení mohou být velmi časově a technicky náročné. Nicméně současné použití pasivních vzorkovačů, chemické analýzy a biotestů se ukázalo jako užitečný přístup pro charakterizaci znečištění odpadní vody, toho, jaké relativní riziko představuje a jaké je jeho bezpečné zředění po vypuštění do prostředí. Tato disertace jako celek prezentuje moderní přístup k hodnocení toxicity s využitím embryonálního testu s Danio rerio s cílem poskytnout spolehlivá data o toxicitě a zároveň mechanistické informace, které splňují požadavky environmentálního a chemického hodnocení rizik 21. století.

Abstract:

The environmental pollutants of anthropogenic and natural origin are an ongoing problem in the aquatic environment and their successful risk management relies on toxicity data. There is a demand for more information and data on the hazards that an increasing number of compounds might pose while there is a strong push to suppress toxicity testing on animals to a minimum. That results in relying more on alternative approaches, in silico and in vitro methods and other alternatives that replace testing using adult animals. This thesis specifically focuses on the use of the zebrafish (Danio rerio) embryo model, small Cyprinidae fish with a number of advantages for toxicity testing. The general objective of this thesis is to develop and apply new approaches to study the effects of environmental pollutants with focus on the aquatic environment and fishes and the mechanisms underlying the adverse outcomes that might occur in the environment. The zebrafish embryo assay was used to characterize the toxicity of anthropogenic aquatic pollutants like pesticides and mixtures like wastewaters or cyanobacterial extracts. We have employed novel approaches to study the teratogenicity of cyanobacterial blooms extracts in combination with detailed mixture content characterization and fractionation and in vitro testing for the retinoid receptor-mediated activity to identify the specific compounds contributing to the effects (Study I and II). We have found that the fractions of the mixture with retinoid compounds are responsible for the vast majority of the toxicity. Based on our results we have established a link between the retinoid compounds present in the cyanobacterial blooms extracts and the teratogenicity that occurs in vertebrates. We also detected neurotoxic effects in fractions with retinoids, but those we were not able to link with specific compounds and the role of retinoids remains unclear. To study the effects of the widely used pesticides we have employed extended zebrafish embryo test to address various effects on organs and included mechanistic endpoints like monitoring of cardiotoxicity (Study III and V). We have obtained valuable results for effects on molecular biomarkers after exposure to environmental concentrations. We showed that zebrafish embryo assay represents a powerful tool for chemical environmental risk assessment since it can provide mechanistic data besides the standard endpoints like mortality and malformations that are increasingly asked for at the European regulatory level. We have also used the zebrafish embryo assay with extended endpoints for toxicity characterisation of complex produced waters complemented with extensive chemical analysis (Study IV). We have shown that bioassay testing is a powerful tool for mixture toxicity assessment since the chemical analysis and subsequential effect estimation based on the content can prove exhaustive. However, complimentary use of passive samplers, chemical analysis and bioassay showed as a useful approach how to monitor and assess the waste-water pollution, the relative risk it poses and the safe dilution factor after the produced water is discharged. This dissertation presents a modern approach to toxicity assessment by utilization of the zebrafish embryo assay to provide reliable toxicity data as well as mechanistic information to fulfil the needs of environmental and chemical risk assessment of the 21st century.
