Determination Of Minor Fatty Acids At Plasma Of Patients With Adrenoleukodystrophy

Abstract:

Diplomová práce je zaměřena na problematiku diagnostiky adrenoleukodystrofie. Toto onemocnění se vyznačuje defektem metabolismu lipidů a peroxizomální B-oxidace, které následně způsobují akumulaci mastných kyselin s velmi dlouhým řetězcem v plazmě a tkáních pacientů, především pak v mozku a nadledvinkách. K analýze byla nejprve použita metoda tenkovrstvé chromatografie, pomocí které byly jednotlivé vzorky rozděleny do pěti lipidových frakcí. Následně byla stanovena koncentrace jednotlivých mastných kyselin ve všech frakcích pomocí plynové chromatografie s plamenově ionizačním detektorem. V této diplomové práci bylo celkem analyzováno deset vzorků plazmy pacientů, trpících adrenoleukodystrofií, a jedenáct kontrolních vzorků zdravých jedinců. Biochemická laboratorní diagnostika adrenoleukodystrofie spočívá především ve stanovení koncentrace mastných kyselin s velmi dlouhým řetězcem a jejich poměrů. Cílem této práce však bylo zaměřit se na tzv. minoritní mastné kyseliny a určit, zda by se hodnoty koncentrací těchto mastných kyselin daly využít jako další biochemický marker adrenoleukodystrofie. Na základě získaných dat bylo prokázáno, že stanovení některých minoritních mastných kyselin by mohlo být využíváno jako podpůrný diagnostický marker adrenoleukodystrofie.

Abstract:

The diploma thesis is focused on the issue of diagnosis of adrenoleukodystrophy. This disease is characterized by a defect in lipid metabolism and peroxisomal B-oxidation, which subsequently cause the accumulation of very long-chain fatty acids in the patient's plasma and tissues, primarily in the brain and adrenal glands. For the analysis, the thin-layer chromatography method was used, when the individual samples were divided into five fractions. Subsequently, the concentration of individual fatty acids in all fractions was determined by gas chromatography with a flame ionization detector. In this thesis, ten plasma samples of patients suffering from adrenoleukodystrophy and eleven control samples of healthy individuals were analyzed. The biochemical laboratory diagnosis of adrenoleukodystrophy primarily determines the concentration of very long-chain fatty acids and their ratios. However, the goal of this work was to focus on so-called minor fatty acids and to determine, whether the concentration values of these fatty acids could be used as other biochemical markers of adrenoleukodystrophy. Based on the obtained data, it was determined that the detrmination of some minor fatty acids could be used as a supporting diagnostic marker of adrenoleukodystrophy.
