Response of plant root system to drought stres

Abstract:

Prvním cílem diplomové práce bylo vytvoření kultivačního systému, který by stimuloval nekolísající nízkou dostupnost vody v substrátu (působením polyethylenglykolu-PEG) a srovnání rostlin z tohoto systému s rostlinami, které prošly periodickým vysycháním. Druhým cílem práce bylo zjistit, jak je růst kořenového systému ovlivněn sníženým vodním potenciálem a periodickými změnami v dostupnosti vody v substrátu. Experimentální rostlinou byl zvolen bob obecný (Vicia faba). Rostliny byly pěstované v keramické kultivační drti a zalévané pomocí kapkové závlahy. Nebylo využito hydroponie, aby nedocházelo k ovlivnění morfologie a anatomie kořenů. U rostlin kontrolních, stresovaných PEG a postupně vysychajících byla změřena biomasa, vodní potenciál, rychlost transpirace a kořeny byly vyhodnoceny v programu Smartroot. Nejprve bylo nutné stanovit optimální hodnotu vodního potenciálu roztoku s PEG. Po experimentech s -0,9 MPa a – 0,6 MPa, byl stres snížen na -0,4 MPa. I přes toto snížení byly rostliny ovlivněné PEG více stresované v porovnání s rostlinami postupně vysychajícími. Příkladem může být biomasa, která byla u rostlin ovlivněných PEG téměř poloviční oproti rostlinám, které postupně vysychaly. Rozdíl v působení stresů mohl být také způsoben nedokonalostí kultivačního systému s kapkovou závlahou.

Abstract:

The first aim of this thesis was to create a cultivation system which stimulates the irregular low availability of water in the substrate (by the action of polyethylene glycol - PEG) and the comparison of plants from this system with plants that have undergone periodic drying. The second aim of the thesis was to find out how the growth of the root system is influenced by the reduced water potential and periodic changes in the availability of water in the substrate. As the experimental plant was selected a bean (Vicia faba). Plants were grown in ceramic cultured crushed and drip irrigation system. Hydroponics was not used to avoid influencing of morphology and roots anatomy. For controlled, PEG stressed and gradually drying plants, biomass, water potential, transpiration rate were measured and roots were evaluated in the Smartroot program. First, it was necessary to determine the optimal water potential of the PEG solution. After an experiment with -0.9 MPa and -0.6 MPa, the stress was reduced to -0.4 MPa. Despite this reduction, plants affected by PEG were more stressed compared to plants gradually drying out. For example, biomass was 50 % lower in plants affected by PEG in comparison to plants that gradually dried out. The difference in the effects of stress can also be due to the imperfection of the drip irrigation system
