Induction of heat disipation of excitation energy in photosystem II during short term exposure of choosen plant species to elevated temperatures and excess irradiance

Abstract:

Bakalářská práce je zaměřena na studium vlivu zvýšené teploty a nadměrné ozářenosti na indukci tepelné disipace excitační energie ve fotosystému II. V teoretické části jsou popsány mechanismy poškození fotosyntetického aparátu způsobené vysokou teplotou a nadměrnou ozářeností a procesy, které chrání PSII před poškozením zmíněnými stresory. Hlavním cílem experimentální části bakalářské práce bylo ověřit následující hypotézy: 1) U smrku a Arabidopsis je rozdílná teplotní závislost indukce nezářivé disipace excitační energie (NRD) ve světlosběrných komplexech fotosystému II mezi různými rostlinami; 2) U smrku je ve větší míře urychlen xantofylový cyklus vlivem zvýšené teploty než u Arabidopsis thaliana. Měření byla provedena na jehlicích smrku ztepilého a listech Arabidopsis thaliana (divoký typ, WT) kultivovaných při optimálních podmínkách. Fluorimetr PAM 101-103 byl využit k měření fluorescence chlorofylu a při různých teplotách (20°C, 32°C, 36°C a 40°C), intenzitě světla 1400 ?mol.m-2.s-1 a světelných režimech (1s, 4s, 7s, 10s, 10x1s a 600s). Z výsledků byly vypočítány následující parametry: potenciální kvantový výtěžek fotochemické reakce PSII (FV/FM), míra NRD ve světlosběrných komplexech fotosystému II (SV0). Výsledky prokázaly, že u smrku dochází ke stimulaci rychlé fáze NPQ již při teplotě 32°C. Naopak u Arabidopsis thaliana se indukuje rychlé zhášení fluorescence pouze při teplotě 40°C. Smrk ztepilý a Arabidopsis thaliana tedy vykazují odlišnou teplotní závislost indukce zhášení fluorescence. Po opakovaných 1s pulsech dochází k rychlé indukci zhášení fluorescence F0 již při teplotě 32°C u smrku ztepilého. Analýza dynamiky světlem indukované deepoxidace violaxantinu ukázala, že došlo k urychlení deepoxidace violaxantinu na zeaxantin přes anteraxantin a akumulaci zeaxantinu pouze u jehlic smrku. U Arabidopsis dochází k průkazné změně odezvy fluorescence chlorofylu a na saturační pulsy až při teplotě 40°C. Místo rychlého poklesu fluorescence pod F0 je po vypnutí světla pozorován výrazný postiluminační nárůst fluorescence pravděpodobně vlivem zrychleného cyklického transportu elektronů kolem PSI a je urychlena pouze první reakce deepoxidace violaxantinu na anteraxantin. U smrku se tedy ve větší míře podílí na ochraně PSII NRD závislá na zeaxantinu. Během 10 minutového osvětlení o intenzitě 1400 ?mol.m-2.s-1 se při teplotě 20°C indukovala NRD ve světlosběrných komplexech PSII u obou rostlin. Při teplotě 40°C byly naměřeny nižší hodnoty SV0 a u Arabidopsis se indukovalo NRD pouze po 1. minutě. Zvýšená teplota tedy nezpůsobuje zvýšení hodnoty SV0.

Abstract:

The bachelor thesis is focused on the study of effects of elevated temperature and excess irrandiance on the induction of heat dissipation of excitation energy within photosystem II (PSII). In the theoretical part mechanisms of photosynthetic apparatus damage caused by high temperature and excess irradiance and processes that protect the photosystem II against these stressors are described. The main goal of experimental part of presented bachelor thesis was to evaluate the following hypotheses: 1) Different temperature dependence of rapid induction of non-radiative dissipation of excitation energy within light-harvesting complexes of photosystem II among different plant species; 2) In spruce needles the xanthophyll cycle due to elevated temperature can be accelerated to higher degree than in Arabidopsis thaliana leaves. The experiments were carried out on spruce needles and A. thaliana leaves (wild type, WT) cultivated under optimal conditions. Chlorophyll a fluorescence were measured by fluorometer PAM 101-103 at different temperatures (20°C, 32°C, 36°C and 40°C), saturation light intensity 1400 ?mol.m-2.s-1 and illumination regimes (1s, 4s, 7s, 10s, 10x1s, 600s). The following chlorophyll fluorescence parameters were calculated: the potential quantum yield of PSII photochemistry (FV/FM), the rate of nonradiative dissipation (NRD) of excitation energy within photosystem II (SV0). Results revealed that the rapid NRD of excitation energy within photosystem II is induced already at 32°C in spruce needles, but only at 40°C in A. thaliana leaves. Norway spruce and A. thaliana have different temperature dependence of induction of NRD of excitation energy within photosystem II. The rapid quenching of F0 fluorescence after 1s light pulses can be observed in spruce needles already at 32°C. The analysis of dynamics of light-induced violaxanthin deepoxidation revealed that deepoxidation of violaxanthin to zeaxanthin via an antheraxanhin was accelerated and the accumulation of zeaxanthin was observed only in spruce needles. In Arabidopsis leaves the chlorophyll fluorescence response to saturation pulses was changed significantly only at 40°C. Instead of rapid quenching below the F0 the pronounced post-illumination fluorescence rise probably due to enhanced cyclic electron flow around photosystem I was observed. Moreover we have found that in A. thaliana leaves only the first step of deepoxidation to antheraxanthin was accelerated. Zeaxanthin-dependent non-radiative dissipation of excitation energy (NRD) is involved to a higher extent in PSII protection in spruce needles than in A. thaliana leaves. NRD within LHCII was induced in both plants during 10 minute illumination at 20°C. The lower values of SV0 were measured at 40°C and NRD was induced only after first minute of illumination in A. thaliana leaves. It follows that the value of SV0 doesn´t increase with the elevated temperature.
