Identification of amino acid residues of YiuR protein recognizing colicin FY

Abstract:

Colicin FY is just recently discovered antimicrobial substance. The toxin is produced by non pathogenic strain Yersinia frederiksenii Y27601. The effect of the colicin FY on other microorganisms is based on the presence of protein YiuR on the outer membrane of the bacteria. Colicin FY acts against bacteria of the genus Yersinia, mainly the pathogenic strain Y. enterocolitica where the resistance has not been observed yet. On the other hand, another gastrointestianl pathogen Y. pseudotuberculosis is fully resistant toward this colicin. Highly specific interactions of the colicin with the receptor are responsible for the specificity of the colicin FY. That is why it was suggested to study variable sequences of the receptor protein YiuR of the colicin sensitive and colicin resistant yersinias. The thesis is based on molecular biology methods such as site-directed mutagenesis, cloning, sequencing and following detection of the colicin impact on newly created recombinant strains.

Abstract:

Kolicin FY představuje antimikrobiální látku produkovanou nepatogenním kmenem Yersinia frederikseni Y27601. Účinek kolicinu FY na jiné mikroorganizmy je závislý na přítomnosti proteinu YiuR (Yersinia iron uptake receptor) na vnější membráně. Kolicin FY je účinný na mikroorganizmy rodu Yersinia a to především na patogenní kmen Y. enterocolitica, u nějž do této doby rezistence nebyla zjištěna. Naproti tomu stojí další gastrointestinální patogen Y. pseudotuberculosis, který je plně rezistentní k účinkům tohoto kolicinu. Rezistence a senzitivita jsou podmíněné vysoko specifickými interakcemi kolicinu s receptorem. Cílem diplomové práce je studium variabilních sekvencí receptorového proteinu YiuR u kolicin-senzitivních a kolicin-rezisentních yersinií s využitím metod molekulární biologie jako cílená mutageneze, klonování a sekvenování s následnou detekcí účinku kolicinu na vzniknuté rekombinantní kmeny.
