Antiviral effect of synthetic derivatives of nucleotides and nucleosides against DNA and RNA viruses

Abstract:

Práce se zabývá látkami nukleotidového a nukleosidového charakteru, které vykazují antivirální účinek. Dále je v práci pojednáno o liposomálních nosičích, které jsou vhodným nanopartikulárním systémem pro cílení léčiv včetně antivirotik. Herpesviry a flaviviry představují významnou skupinu lidských a zvířecích patogenů. Proti naprosté většině flavivirů význačných z medicínského hlediska nejsou komerčně dostupná antivirotika a schválená terapie. Úspěšná antivirová léčba je dostupná proti řadě herpesvirů. V případě Aujeszkyho choroby však doposud nebyla schválena žádná účinná chemoterapie. Značnou komplikaci v léčbě virových infekcí představuje vznik rezistentních forem. Z těchto důvodů je cílem předkládané práce nalézt vhodné látky pro inhibici virové infekce. Nukleosidové analogy jsou základními terapeutiky pro léčbu virem infikovaných pacientů, proto byla vybrána série látek z této skupiny pro testování antivirálního účinku proti zvoleným herpesvirovým a flavivirovým patogenům. V případě herpesvirů byly zjištěny dva účinné nukleosidové fosfonáty (9-[2-(phosphonomethoxy) ethyl]-2,6-diaminopurin a N6-cyclopropyl-9-[2-(phosphonomethoxy) ethyl]-2,6-diaminopurin) se značným inhibičním účinkem in vitro proti viru Aujeszkyho choroby. Dále byly nalezeny významné inhibitory flavivirové infekce, a to imino-C-nukleosid BCX4430, kde byl antivirový účinek prokázán in vitro proti viru klíšťové encefalitidy (VKE), západonilské horečky, horečky Kyasanurského lesa a vrtivky a 7-deaza-2´-C-methyladenosin (7-deaza-2´-CMA), který silně inhiboval VKE i v myším modelu. Mechanismus inhibičního účinku 7-deaza-2´-CMA byl rovněž studován prostřednictvím mutant VKE rezistentních na 2´-C-methylované nukleosidy. Tyto mutanty jsou charakteristické substitucí S603T, která je zodpovědná za vysoký stupeň rezistence na 2´-C-methylované nukleosidy a současně udává viru silně atenuovaný fenotyp, který vede ke snížené virulenci viru na myším infekčním modelu. Pro cílení antivirotik lze využít liposomy, které představují významné netoxické biokompatibilní nanočástice používané především pro cílený transport léčiv. Aktivního a specifického cílení liposomálních nosičů je dosaženo modifikací povrchu liposomů s využitím různých ligandů, jako jsou sacharidy a polysacharidy. Byla vyvinuta metoda přípravy nanoliposomů modifikovaných kyselinou hyaluronovou s využitím mikrofluidního mísení a klik chemie na bázi oximové ligace. Pro tento účel byly vyvinuty nové aminooxy lipidy.

Abstract:

The thesis deals with compounds of nucleotide and nucleoside character, which have an antiviral effect. Furthermore, the thesis deals with liposomal carriers, a suitable nanoparticular drug targeting system including antivirals. Herpesviruses and flaviviruses represent a group of medically important pathogens of humans and animals. There are no available antivirals and approved therapy against most flaviviruses. A successful antiviral treatment is available against a number of representatives of the Herpesviridae family. No effective chemotherapy has been approved for Aujeszky’s disease so far. A major complication in the treatment of viral infections is the emergence of drug resistant forms. For these reasons, the aim of the present thesis is to find suitable agents for inhibiting viral infections. Nucleoside analogues are basic therapeutic for the treatment of virus infected patients, therefore a series of compounds from this class has been chosen to evaluate their antiviral activity against selected herpesvirus and flavivirus pathogens. In the case of herpesviruses, two effective nucleoside phosphonates (9-[2-(phosphonomethoxy) ethyl]-2,6-diaminopurine and N6-cyclopropyl-9-[2-(phosphonomethoxy) ethyl]-2,6-diaminopurine) were found to show a significant inhibitory activity against Aujeszky’s disease virus in vitro. In addition, strong inhibitors of flavivirus infection were found. An imino-C-nucleoside BCX4430 exerted an in vitro antiviral activity against tick-borne encephalitis virus (TBEV), West Nile virus, Kyasanur Forest disease virus and louping ill virus, whereas 7-deaza-2´-C-methyladenosine (7-deaza-2´-CMA) displayed a micromolar inhibitory effect against TBEV in mouse model. The mechanism of inhibitory activity of 7-deaza-2'-CMA has also been studied by TBEV mutant resistant to 2'-C-methylated nucleosides. These mutants are typical the S603T substitution, which is responsible for a high-level resistance to 2'-C-methylated nucleosides. Interestingly, the mutant virus showed a significantly attenuated phenotype compared with the wild-type virus, which was manifested by a reduced virus virulence in the mouse infective model. Liposomes, significant non-toxic biocompatible nanoparticles used primarily for a targeted drug transport, can be used for targeting antivirals. Active and specific targeting of liposomal carriers can be achieved by modifying the surface of liposomes using various ligands such as saccharides and polysaccharides. A method for the preparation of nanoliposomes modified with hyaluronic acid has been developed using microfluidic mixing and click-chemistry based on oxime ligation. New aminooxy lipids have been developed for this purpose.
