Effect of cultivation conditions on transcriptome of bacteria Streptococcus mutans

Abstract:

Streptococcus mutans je kvůli své schopnosti metabolizovat exogenní sacharidy za současné produkce organických kyselin považován za hlavního původce zubního kazu. Cílem této diplomové práce bylo analyzovat změny v transkriptomu kariogenní bakterie Streptococcus mutans zapříčiněné dostupností různých sacharidů v kultivačním médiu. Součástí této práce byla izolace RNA z bakteriálních kultur, příprava sekvenační knihovny a bioinformatické zpracování sekvenačních dat za účelem porovnání genové exprese a metabolických drah. U vzorků kultivovaných na glukóze a sacharóze byla pozorována represe galaktózového metabolismu, která je v literatuře popsána jako důsledek CCR mechanismu. U vzorků kultivovaných na xylitolu docházelo u bakterie k hladovění a byla pozorována aktivace metabolismu pyruvátu a acetyl-CoA, při níž si bakterie přes intermediát acetyl-P generovala acetát a ATP. Naše výsledky se z velké části shodují s dřívějšími experimenty podobného typu a přispívají k objasnění metabolismu kariogenního S. mutans.

Abstract:

Streptococcus mutans is considered to be a major cause of tooth decay due to its ability to metabolize exogenous carbohydrates with the simultaneous production of organic acids. The aim of this diploma thesis was to analyze changes in the transcriptome of the cariogenic bacterium Streptococcus mutans caused by the availability of various carbohydrates in the culture medium. Part of this work was the isolation of RNA from bacterial cultures, preparation of a sequencing library and bioinformatics processing of data in order to compare gene expressions and metabolic pathways. In samples cultured on glucose and sucrose, repression of galactose metabolism has been observed, which is described in the literature as a consequence of the CCR mechanism. In samples cultured on xylitol, the bacterium starved and activation of pyruvate and acetyl-CoA metabolism was observed, in which the bacterium generated acetate and ATP via the acetyl-P intermediate. Our results are largely consistent with previous experiments of a similar type and contribute to the elucidation of the metabolism of cariogenic S. mutans.
