Differences in morphology, tissue structure, and phenolic compound distribution in plants induced by light of varying spectral quality

Abstract:

V rámci bakalářské práce byl sledován vliv spektrální kvality fotosynteticky aktivního záření na morfologii, anatomii a distribuci fenolových látek na dvou modelových rostlinách - ječmeni setém (Hordeum vulgare) a huseníčku rolním (Arabidopsis thaliana). V případě huseníčku byl využit nejen divoký typ (Landsberg erecta - Ler-0, Columbia - Col-0), ale i pět mutantních linií - s mutací v genu kódujícím hydroxylázu kyseliny ferulové (FAH1) (byly použity dvě různé linie), s mutací v genu kódujícím chalkonsyntázu (tt4), dvojitý mutant (FAH1tt4) a se značenými subcelulárními strukturami (kaleidocell). Oba druhy rostlin byly prekultivovány na bílém světle při nízké ozářenosti 100 umol m-2 s-1 a následně aklimovány na různé spektrální kvality při ozářenosti 400 umol m-2 s-1. Ječmen byl aklimován na červené, modré a zelené světlo, včetně jejich dichromatických kombinací, a huseníček byl aklimován na bílé, modré a červeno-zelené světlo. Bylo posuzováno, jak spektrální kvalita ovlivňuje výšku rostlin, délku listů, plochu prýtu i listu, čerstvou hmotnost i hmotnost sušiny. Na mikroskopické úrovni byla pomocí světelné mikroskopie sledována šířka listu a mohutnost palisádového parenchymu. Pomocí epifluorescenční mikroskopie byla hodnocena distribuce fenolových látek na příčném řezu listem. Kromě toho byla nově zavedena metodika fluorescenčního barvení flavonoidů na příčném řezu pomocí Naturstoff reagentu. Většina sledovaných parametrů spojených s rychlostí růstu rostlin a listů (výška prýtů, délka listů, plocha listů, čerstvá hmotnost listů) byla modrým světlem, oproti ostatním spektrálním kvalitám, negativně ovlivněna. Tento negativní vliv je v souladu s výsledky uvedenými v odborné literatuře a je zřejmě zapříčiněn aktivací kryptochromů na modrém světle. Avšak tato modrým světlem aktivována signální dráha vede k výrazně vyšší akumulaci fenolových látek v listech, přičemž tato práce blíže specifikuje právě jejich distribuci v rámci pletiv listů. Tato odezva je pravděpodobně jedním z aklimačních procesů souvisejících se zvyšováním odolnosti vůči fotooxidativnímu stresu.

Abstract:

The presented bachelor thesis studied the impact of the spectral quality of photo-synthetically active radiation on plant morphology, anatomy, and distribution of phenolic compounds in two different model plant species - specifically on spring barley (Hordeum vulgare), and thale cress (Arabidopsis thaliana). Except the Arabidopsis thaliana wild type (Landsberg erecta - Ler-0, Columbia - Col-0), five different mutant lines were used - with mutation in the gene encoding ferulic acid hydroxylase (FAH1) (two different mutant lines were used), with mutation in the gene encoding chalcone synthase (tt4), a double mutant (FAH1tt4) and with labelled subcellular structures (kaleidocell). Plants were precultivated under white light and low irradiance (100 umol m-2 s-1) and then acclimated to various spectral qualities and higher irradiance (400 umol m-2 s-1). Barley was acclimated to red, blue and green light, including their dichromatic combinations, and thale cress was acclimated to white, blue and red-green light. It was determined, how the spectral quality affects the height of plants, length of leaves, area of shoots, leaf area, fresh weight, and dry weight. The leaf width and thickness of palisade parenchyma were evaluated by use of light microscopy technique. The distribution of the phenolic compounds within the leaf cross-sections was assessed by using epifluorescent microscopy. In addition, a new methodology for fluorescence staining of flavonoids on transverse leaf sections using Naturstoff reagent was introduced. Most of the measured parameters associated with plant and leaf growth rate, such as shoot height, leaf length, leaf area, and leaf fresh weight, exhibited negative response when exposed to blue light in comparison to other spectral qualities. This negative effect is consistent with results reported in the literature and is probably caused by the activation of cryptochromes under blue light. However, activated cryptochromes lead to a significantly higher accumulation of phenolic compounds in leaves, and this work further specifies their internal localization. This response is likely to be one of the acclimation processes associated with increasing resistance to photooxidative stress.
