Sample Preparation for Imaging Mass Spectrometry of Tissues using MALDI MS

Abstract:

Tato práce v teoretické části shrnuje poznatky o zobrazovací hmotnostní spektrometrii s laserovou desorpcí a ionizací za účasti matrice, o přípravě vzorků a metodách nanášení roztoků matrice na vzorky dvěma metodami – elektrosprejem a sprejem. Experimentální část je zaměřená na nanášení roztoku matrice na modelové vzorky těmito dvěma metodami. Pro nanášení jsou sestaveny aparatury, obě metody jsou optimalizovány a následně zkoumány z hlediska náročnosti procesu nanášení a kvality nanesené matrice.

Abstract:

This thesis summarizes the knowledge of matrix-assisted desorption/ionization mass spectrometry imaging, sample preparation and methods of matrix solution deposition over samples in the theoretical part. Two methods are used – electrospray and spray. Experimental part focuses on matrix solution deposition over the model samples using these two methods. Apparatuses for deposition have been built. Both methods are optimized and further investigated with regard to the process deposition feasibility and matrix deposition quality.
