Arrayed nanostructures for electrochemical sensors

Abstract:

Předkládaná disertační práce se zabývá dnes velmi aktuálním tématem a to vytvářením a charakterizací nanostruktur. Nanostrukturované materiály se stávají stále více středem pozornosti mnoha vědeckých týmů v různých oblastech vědy a techniky právě díky svým unikátním vlastnostem. V rámci této disertační práce se jedná především o zvětšení aktivní plochy elektrod nanostrukturováním povrchů a také možnost imobilizace biologicky důležitých látek na vytvářených nanostrukturách. Pro experimentální zpracování disertační práce bylo využito elektrolytického vylučování kovu do nanoporézní nevodivé masky, což je přístrojově i finančně relativně nenáročná metoda. Nanostruktury se podařilo vytvořit ze tří různých kovů a to niklu, zlata a mědi. Seskupené nanostruktury byly následně analyzovány rastrovacím elektronovým mikroskopem a u vybraných vzorků energiově disperzní rentgenovou spektroskopií. Pro další charakterizaci bylo v případě měděných nanostruktur využito také elektrochemických metod, konkrétně elektrochemické impedanční spektroskopie a diferenční pulzní voltametrie. V neposlední řadě byly měděné seskupené nanostruktury využity k elektrochemické detekci adeninu pomocí již zmíněné voltametrické techniky. Byla zjišťována využitelnost nanostrukturovaného měděného povrchu jako elektrody pro potenciální aplikace v oblasti senzorů a biosenzorů. Výsledky získané z měření pomocí nanostrukturovaných povrchů byly také srovnávány s povrchy bez nanostruktur a byly hledány vhodné podmínky pro detekci purinové báze adeninu. Disertační práce přispívá svými výsledky do oblasti senzoriky a poskytuje podklad pro další vývoj moderních biosenzorů. Práce je rozdělena do teoretické a praktické části. Teoretická část se zabývá poznatky již dosažitelnými ve vědecké literatuře a tématem celkově na teoretické úrovni. Část praktická navazuje na poznatky z literatury a popisuje výsledky depozice kovu z elektrolytu do nanoporézní masky, ukazuje vliv různých parametrů elektrodepozice na výsledné nanostruktury, charakterizuje nanostruktury pomocí ekvivalentních obvodů navržených na základě spekter naměřených v elektrochemické impedanční spektroskopii a naznačuje detekční možnosti nanostruktur jako elektrod ve voltamterických technikách.

Abstract:

The submitted thesis deals with the fabrication and characterization of nanostructures which are currently at the centre of attention of many scientific groups. The nanostructured materials exhibit unique properties in comparison to bulk materials. For example, biologically important species can be immobilized on the nanostructures and also the area of the nanostructured electrode is larger in comparison to the flat electrode. Therefore, nanostructured surfaces are very promising in the field of sensing and biosensing. The fabrication of nanostructures consisted in template-assisted metal deposition from electroplating baths, which is rather simple and inexpensive way to create an array of nanostructures. The nanostructures were then examined using scanning electron microscopy, eventually energy dispersive x-ray spectroscopy. In the case of copper nanostructures, electrochemical impedance spectroscopy and differential pulse voltammetry were also used for further characterization. Differential pulse voltammetry was employed for experiments with adenine detection. Obtained results were than compared with those measured using flat electrodes. Therefore, the main contribution of the thesis lies in the field of biosensing and provides research background for development of advanced biosensors. The thesis is divided into theoretical and practical part. The theoretical part is focused on the existing information available in scientific literature and deals with the subject on theoretical ground. The practical part describes the results of electroplating into membranes, shows the effect of various electroplating parameters on final nanostructures, characterizes nanostructures using equivalent circuits based on electrochemical impedance spectra, and indicates potential applications for electrochemical detection of adenine on copper nanostructures.
