Metal nanoparticles for enhanced response of biosensors for phenols

Abstract:

Bakalářská práce se zaobírá vylepšením biosensorů pro fenoly využitím nanočástic zlata, platiny a redukovaného oxidu grafénu. Zmíněné nanočástice disponují s řadou prospěšných vlastností na zlepšení vodivosti, elektrických vlastností, zvětšují reakční povrch a pomáhají při imobilizaci enzymu. Cílem práce bylo vylepšení odezvy a meze detekce biosensorů pro fenoly využitím elektrod modifikovaných nanočásticemi. Detekce fenolů se prováděla pomocí enzymu tyrosináza, která štěpí mono- a 1,2-difenoly na elektroaktivní chinoidní produkty. Amperometrické stanovení probíhalo na čtyřech typech elektrod a na dvou modelových substrátech (pyrokatechol, fenol) s koncentrací od 1 mM až do nejnižší detekovatelné koncentrace. Ze získaných údajů vyplívá několikanásobné vylepšení odezvy a řádové vylepšení meze detekce elektrodami modifikovanými nanočásticemi. Nejlepší výsledky na substrát pyrokatechol byly dosáhnuty zlatými nanočásticemi a při substrátu fenol platinovými nanočásticemi.

Abstract:

The purpose of this bachelor thesis is to achieve enhanced response of biosensors for phenols with the addition of gold, platinum and reduced graphene oxide nanoparticles. These nanoparticles possess series of beneficial properties to enhance conductivity, electrical properties, increasing the reaction surface and provide support for the enzyme immobilization. The aim of this thesis was to enhance the response and detection of biosensors for phenols with electrodes modified with nanoparticles. The detection of phenols was conducted with the enzyme tyrosinase, which reacted with mono- and 1,2-diphenols to produce electroactive quinones. The amperometric detection was carried out on four different types of electrodes with two model substrates (pyrocatechol, phenol) with the concetration from 1 mM to the lowest detectable limit. The acquired data show significant increase in response and detection on the electrodes modified with nanoparticles. The best results for the substrate pyrocatechol were achieved by gold nanoparticles and for phenol by platinum nanoparticles.
