Mechanisms of ring transformations of isothiouronium salts derived from bromolactones

Abstract:

The series of isothiouronium salts was prepared by the reaction of 2-bromobutyrolactone with substituted thioureas. Products of their transformation in aqueous solutions were characterized. The kinetics and mechanism of rearrangement of S-(2-oxotetrahydrofuran-3-yl)-N-(4-methoxyphenyl)isothiouronium bromide into 5-(2-hydroxyethyl)-2-[(4-methoxyphenyl)imino]-1,3-thiazolidin-4-one have been studied in aqueous buffer solutions. Multiple breaks in the measured pH profile establish the formation of three different kinetically detectable intermediates T?, T0 and T-, whose rate of formation and breakdown to the product are pH dependent. Influence of the additional Cu(II) ions on the reaction rate has been followed. In the second part of the work 3-bromophtalide have been used instead of the 2-bromobutyrolactone. This alteration causes the change in the mechanism and reaction leads to the different types of the products depending on structure.

Abstract:

Série isothiouroniových solí byla připravena reakcí 2-brombutyrolaktonu se substituovanými thiomočovinami. Produkty transformace těchto solí ve dvodných roztocích byly charakteryzovány. Transformační reakce S-(2-oxotetrahydrofuran-3-yl)-N-(4-methoxyfenyl)isothiouronium bromidu na 5-(2-hydroxyethyl)-2-[(4-methoxyfenyl)imino]-1,3-thiazolidin-4-on byla sledována spektrofotometricky za podmínek pseudoprvního řádu. pH Profil reakce ukazuje několik zlomů, jejichž přítomnost lze vysvětlit uplatněním tří různých intermediátů T?, T0 a T-. Rychlost jejich vzniku a rozpadu závisí na pH prostředí. Také byl pozorován vliv přídavku měďnatých iontů na rychlost reakce. V další části práce byla obměněna struktura laktonové části molekuly a 2-brombutyrolakton byl nahrazen 3-bromftalidem. Byl pozorován velký vliv substituce isothiouroniové části molekuly na reaktivitu solí a typ vznikajících produktů.
