The role of centrosomes in tumorigenesis

Abstract:

Centrosom je malá organela umístěná v blízkosti buněčného jádra. Tato organela má funkci mikrotubuly organizujícího centra, které kontroluje počet, umístění a orientaci mikrotubulů v buňce. Každá buňka obsahuje během mitózy dva centrosomy, které vytvářejí bipolární dělící vřeténko a tím zajišťují vyvážený přenos chromosomů do dceřiných buněk během mitózy. V případě deregulace duplikace centrosomu dochází k amplifikaci centrosomu, která vede k výskytu multipolárního dělicího vřeténka. Abnormální dělicí vřeténko pak způsobuje chybný přenos chromosomů do dceřiných buněk, což nakonec vede k aneuploidii, která je charakteristickým znakem nádorově transformovaných buněk. Cílem této bakalářské práce je charakterizovat centrosomy, jejich strukturu a funkci v nenádorových buňkách, dále vysvětlit mechanismus duplikace centrosomu a věnovat se možným příčinám poruch, které následně vedou k amplifikaci centrosomu v nádorově transformovaných buňkách. V bakalářské práci je také popsáno, jak amplifikace centrosomu souvisí s chromosomovou nestabilitou v nádorově transformovaných buňkách.

Abstract:

Centrosome is a small organelle found at the periphery of cell nucleus. This organelle functions as a microtubule organizing center controlling amount, position and orientation of microtubules in the cell. During mitosis, each cell contains two centrosomes that make bipolar mitotic spindle in order to accurate movement of chromosomes into daughter cells during mitosis. Deregulation of the centrosome duplication cycle results in abnormal centrosome amplification leads to a formation of aberrant multipolar spindle. Such multipolar spindle causes errors in chromosome movement; resulting aneuploidy of daughter cell is a characteristic feature of tumor cells. The aim of this bachelor thesis is to characterize structure and function of centrosomes in normal cells, to explain the mechanism of centrosome duplication and to address possible causes of its failures, which lead to the centrosome amplification in tumor cells. A relationship between centrosome amplification and chromosomal instability of tumor cells is also described in this bachelor thesis.
