Abstract:

V této disertační práci se zabýváme aplikacemi stochastických bodových procesů v oblasti početních neurověd. V několika úvodních kapitolách jsou připomenuty základní pojmy a metody z teorie pravděpodobnosti a matematické statistiky a představeny vybrané stochastické procesy. V následující kapitole jsou shrnuty nejdůležitější poznatky o fungování neuronů. Ústřední část disertační práce je tvořena pěti odbornými články. Dva z nich se zaměřují na problém odhadu latence odezvy z neuronálních dat. Jsou zde shrnuty a porovnány hlavní metody pro odhad latence excitační odezvy, které lze nalézt v odborné literatuře. Dále uvažujeme problém odhadu latence inhibiční odezvy, kde navrhujeme několik možných metod odhadu, které jsou založeny na pozorování časů prvních neuronálních impulzů po začátku stimulace. V dalších dvou článcích zkoumáme přesnost, s jakou lze odhadnout intenzitu stimulace z latence prvního neuronálního impulzu po začátku stimulace. Uvažujeme několik možných matematických modelů neuronální aktivity (Poissonův proces, proces obnovy, Wienerův proces) a na nich zkoumáme vliv parametrů těchto modelů, především dopad spontánní aktivity neuronů. V posledním přiloženém článku je zkoumána přesnost, s jakou lze intenzitu stimulace odhadnout z frekvence neuronálních impulzů.

Abstract:

This doctoral thesis deals with several applications of stochastic point processes in computational neuroscience. In the first few introductory chapters the basic notions and methods from probability theory and statistics are recalled and several selected stochastic processes are presented. Another chapter summarizes the key knowledge about functioning of neurons. The key part of the thesis consists of five research papers. Two of them focus on the problem of the estimation of response latency from neuronal data. The main methods for estimating the latency of excitatory response, which can be found in literature, are summarized and compared. Then the problem of estimating the latency of inhibitory response is studied and several estimation methods using the observations of first spikes after the stimulus onset are proposed. In the next two papers the accuracy of estimating the stimulus intensity from the first-spike latency is investigated. Different models of neuronal activity are considered (Poisson process, renewal process, Wiener process) and the effect of the key parameters of the models, especially the impact of spontaneous neuronal activity, are analyzed. In the last attached paper we investigate the accuracy, with which the stimulus intensity can be estimated from the spike frequency.
