Gut microbiome in the context of heterogeneity of colorectal cancer

Abstract:

There are many bacteria in the human intestine that are inevitable for the healthy functioning of the organism, and the disturbance of this system is often associated with the development of colorectal cancer (CRC). The advent of next-generation sequential technologies has enabled the characterization of intestinal wetness without the need for a culture medium. This diploma thesis is focused on observation of differences between microbial composition and obtained data from analysis of molecular pathology and immune tumour profile. The results point to differences in tumour microbial structure and normal. Reduced bacterial genera Faecalibacterium and Lachnoclostridium were found in tumour microbiomes in patients with CRC, while the bacterial strain Fusobacterium occurred in a higher percentage. Regarding the presence of the BRAF mutation, reclaimed bacterial genera were identified: Faecalibacterium and Roseburia. In the presence of microsatellite instability, reduced bacterial genera Alistipes, Parabacteroides, Faecalibacterium and Roseburia were found. The work also dealt with the comparison of the microbial composition of the tumour with the immune tumour profile. Reduced bacterial genera Bacteroides and Lachnoclostridium have been identified for elevated PD1. Depending on the increase in PDL-1 exposure, the Bacteroides and Suterella genera were reduced. Bactoroides showed lower levels of incidence even in the presence of increased CD8 expression. Following the above, it can be stated that the results indicate a correlation between the intestinal microflora and the risk of developing CRC. The results also show the possibility of a connection between the molecular and immune tumour profile.

Abstract:

V lidském střevě se nachází množství bakterií, které jsou nevyhnutelné pro zdravé fungování organismu, přičemž narušení tohoto systému je často spjaté i s rozvojem kolorektálního karcinomu (KRK). Příchod sekvenačních technologíí nové generace umožnil charakterizaci střevního mokrobiomu bez nutnosti kultivačního média. Tato diplomová práce je zaměřena na pozorování odlišností mezi mikrobiální kompozicí a získanými daty z analýzy molekulární patologie a imunitního profilu nádoru. Výsledky poukazují na odlišnosti mikrobiální struktury nádoru a normálu. V mikrobiomech nádoru byly u pacientů s KRK zjištěné v menší početnosti bakteriální rody Faecalibacterium a Lachnoclostridium, zatím co bakteriální kmen Fusobacterium se vyskytoval ve vyšším zastoupení. Z hlediska přítomnosti mutace BRAF byly identifikované v nižším zastoupení bakteriální rody: Faecalibacterium a Roseburia. V přítomnosti mikrosatelitní nestability se nacházely v nižším zastoupení bakteriální rody Alistipes, Parabacteroides Faecalibacterium a Roseburia. Práce se věnovala také porovnání mikrobiální kompozice nádoru s imunitním profilem nádoru. Z hlediska zvýšené exprese PD1 byly identifikované v nižším zastoupení bakteriální rody Bacteroides a Lachnoclostridium. V závislosti na zvýšení expese PDL-1 byly redukované rody Bacteroides a Suterella. Rod Bactoroides vykazoval nižší hodnoty výskytu i v přítomnosti zvýšené exprese CD8. V návaznosti na výše uvedené, lze konstatovat, že této výsledky naznačují vzájemnou spojitost mezi střevní mikroflórou a s rizikem vzniku CRC. Výsledky poukazují i na možnost spojitosti mezi molekulárním a imunitním profilem nádoru.
