Morphological and molecular identification of anisakid nematodes from regurgitated pellet of South Polar Skua (<I>Catharacta maccormicki</I>) in James Ross Island, Antarctica

Abstract:

Bakalářská práce je věnována problematice identifikace anisakidních hlístic mořských ptáků v Antarktidě. Anisakidní hlístice byly předběžně identifikovány pomocí morfologických znaků jako „Contracaecum osculatum species complex“. K přesné identifikaci hlístic bylo následně využito metod molekulární biologie. Vzorky byly jednotlivě amplifikovány metodou polymerázové řetězové reakce (PCR) a analyzovány Sangerovou metodou sekvenování. K tomuto účelu byly využity tři molekulární markery: ribozomální DNA (rDNA) - a) ITS-2 (Internal Transcribed Spacer 2), b) gen kódující malou ribozomální podjednotku 18S a mitochondriální DNA (mtDNA) c) COX2 gen pro cytochrom c oxidázu – podjednotka II . Výsledky potvrzují příslušnost hlístic nalezených v trávicím traktu chaluhy antarktické k druhu Contracaecum osculatum. K přesnému určení poddruhu bude zapotřebí provést navazující detailní studie.

Abstract:

Bachelor thesis is devoted to the identification of anisakid nematodes of seabirds in the Antarctic. Anisakid nematodes were tentatively identified using morphological characters, such as „Contracaecum osculatum species complex“. For precise identification of nematodes were then used molecular biology techniques. Samples were individually amplified by polymerase chain reaction (PCR) and analysed by the Sanger sequencing method. Three distinctive molecular markers were used for this purpose: ribosomal DNA (rDNA) – a) ITS-2 (Internal Transcribed Spacer 2), the gene encoding small ribosomal subunit 18S and mitochondrial DNA (mtDNA) c) COX2 gene for cytochrome c oxidase – subunit II. Results confirm the affiliation of nematodes found in the digestive tract of the south polar skua to the species Contracaecum osculatum. To the exact determination of particular subspecies the following detailed study will be needed.
