Regulation of expresion of ephrins and their receptors in haematological malignancies

Abstract:

Ephriny a jejich receptory, včetně receptorů EphA1 a EphA2, jsou známé onkoproteiny. Podporují transformaci buňek a růst nádorů, což je dobře prostudováno u solidních nádorů. EphA2 byl identifikovaný jako ligand pro Vδ1 gamma-delta T buňky a hojné expanze této buněčné populace byly nalezeny u mnohočetného myelomu. Poznatky o expresi a funkci receptoru EphA2 u hematologických malignit však chybí. Prvním cílem diplomové práce byla analýza exprese receptorů EphA1 a EphA2 u nádorových suspesnzních buněčných linií odvozených z hematologických malignit a jejich porovnání s adherentními nádorovými a zdravými buněčnými liniemi. Povrchová exprese obou receptorů byla studována pomocí průtokové cytometrie, množství proteinu EphA2 pomocí Western blotu a exprese genu EPHA2 pomocí kvantitativní real-time PCR. Buněčné linie však představují jen modelový systém, a proto dalším cílem práce byla analýza povrchové exprese EphA2 u B-buněčných malignit, jakými jsou chronická lymfocytární leukemie (CLL), MGUS a mnohočetný myelom (MM). Přestože byly buněčné linie odvozené z MM a CLL EphA2 negativní a leukemické buňky u CLL vykazovaly pouze slabou expresi EphA2, nádorové plasmatické buňky u MGUS a MM byly EphA2 pozitivní. Kromě toho byla povrchová exprese EphA2 detekována u plasmacytoidních dendritických buněk (pDC) u diagnóz MGUS a MM. Více EphA2 pozitivních pDC v kostní dřeni bylo nalezeno u pacientů než u zdravých dárců. Lidské stromální buňky (HS-5) byli také EphA2 pozitivní. Tyto výsledky podporují hypotézu že cytotoxická aktivita Vδ1 gamma-delta T buněk vůči myelomovým buňkám může být podporována interakcemi s receptorem EphA2, který je exprimován u zmíněných buněčných populací v mikroprostrědí kostní dřeně u pacientů s MGUS a MM.

Abstract:

Ephrins and their receptors, including EphA1 and EphA2 receptors, are known as oncoproteins. They promote malignant transformation and tumour growth, which is well studied in solid tumours. The EphA2 was identified as a ligand for the Vδ1 gamma-delta T cells. The large expansions of this cell population were reported in multiple myeloma. However, the knowledge concerning the EphA2 expression and function in haematological malignancies is lacking. The first aim of the thesis was to assess the expression of the EphA1 and EphA2 receptors in tumour suspension cell lines originating from haematological malignancies, compared to the expression in adherent tumour and healthy cell lines. The level of surface expression of the EphA1 and EphA2 was studied by flow cytometry, the EphA2 protein level was analyzed by Western blot and the EPHA2 gene expression by quantitative real-time PCR. Since the cell lines present only a model system, the next aim was to assess the EphA2 surface expression on tumour cells of B cell malignancies, including chronic lymphocytic leukaemia (CLL), MGUS and multiple myeloma (MM). Although the MM and CLL suspension cell lines were uniformly EphA2 negative and leukaemia cells in CLL showed only weak EphA2 expression, tumour plasma cells in MGUS and MM were EphA2 positive. In addition, the EphA2 surface level was determined on plasmacytoid dendritic cells (pDCs) in MGUS and MM. There were more EphA2 positive pDCs in patients’ bone marrow that in healthy donors. Human stromal cells (HS-5) were EphA2 positive as well. These findings support the hypothesis that the cytotoxic activity of Vδ1 gamma-delta T cells reported against malignant cells in MM can be driven by the interactions with the EphA2 receptor expressed on mentioned cell populations within the bone marrow microenvironment of MGUS or MM patients.
