3D Cultivation of Cells

Abstract:

Tato bakalářská práce se zabývá 3D metodami kultivace buněk, jejich základní charakterizací a porovnáním s jinými metodami. V úvodu práce jsou popsány složky extracelulární matrix a buněčná spojení, která mají zásadní vliv na růst buněk v buněčných agregátech. Následně jsou popsány metody buněčné kultivace a jejich základní porovnání ve 2D a 3D systémech. V hlavní části je uveden detailní popis metod 3D kultivace. První skupinou jsou metody bez použití nosiče, kam se řadí metoda visící kapky, kultivace buněk na nepřilnavých substrátech a akustická levitace. Druhou skupinu pak představují metody s využitím nosiče. Do této skupiny se řadí kultivační metody využívající scaffoldy, hydrogely, bioreaktory, biotisk a orgány na čipu. Cílem práce je poskytnout čtenářům hlubší porozumění moderním přístupům v buněčné kultivaci a jejich potenciálu pro biologický a medicínský výzkum.

Abstract:

This bachelor's thesis deals with 3D cell culture methods, their basic characterization and comparison with other methods. The thesis starts with a description of the extracellular matrix components and cell junctions that are essential for cell growth in cell aggregates. Subsequently, cell culture methods and their basic comparison in 2D and 3D systems are described. In the main part, a detailed description of 3D culture methods is given. The first group is the carrier-free methods, which include the hanging drop method, cell culture onnon-adherent substrates and acoustic levitation. The second group is represented by the carrier-assisted methods. This group includes culture methods using scaffolds, hydrogels, bioreactors, bioprinting and organs-on-a-chip. The aim of this work is to provide readers with a deeper understanding of modern approaches to cell culture and their potential for biological and medical research.
