The utilization of MALDI-TOF imaging mass spectrometry for the study of potential markers of animal tissues

Abstract:

MALDI-TOF zobrazovací hmotnostní spektrometrie (MSI) se začala rozvíjet na přelomu druhého a třetího tisíciletí a již od prvních aplikací poskytovala důležité informace o prostorovém výskytu různých biomolekul, nejčastěji peptidů, proteinů, lipidů a oligosacharidů, v řezech živočišných tkání, rostlinných pletiv a podobně. Nejvíce se metoda uplatňuje ve výzkumu potenciálních biomarkerů různých typů nádorů. Tato disertační práce se zabývá využitím MALDI-TOF MSI pro studium prostorové distribuce potenciálních markerů v řezech živočišných tkání. V teoretické části práce je podán obecný přehled o MALDI-TOF hmotnostní spektrometrii a jejím využitím pro zobrazování potenciálních markerů v nádorech. V praktické části práce jsou okomentovány dosažené publikované výsledky. Pomocí MALDI-TOF MSI byla zjištěna prostorová distribuce estradiol 3,4 chinonu, fytochelatinů PC2 a PC3, oxidovaného glutathionu (GSSG) a metalothioneinu v příčných řezech žížaly hnojní (Eisenia fetida). Dále byla tato metoda využita pro mapování metalothioneinu v řezech myších xenograftů nádoru prostaty (PC-3). V případě sledování procesů zahrnutých ve spontánní regresi melanomu u MeLiM (Melanoma-bearing Libechov Minipigs, melanom nesoucí miniaturní prasata z Liběchova) miniaturních prasat byla tato metoda využita pro klasifikaci specifických m/z píků, které signifikantně odlišovaly zdravou kůži a melanom z MeLiM miniaturních prasat.

Abstract:

MALDI-TOF imaging mass spectrometry (MSI) began to develop at the turn of the second and third millennia, and since the first applications it provided important information about the spatial occurrence of various biomolecules, most commonly peptides, proteins, lipids and oligosaccharides, in animal tissue sections. This method has been most widely applied in the research on potential biomarkers of various types of tumors. The dissertation thesis deals with the use of MALDI-TOF MSI for studying spatial distribution of potential markers in animal tissue sections. In the theoretical part of the thesis is provided an overview of MALDI-TOF mass spectrometry and its use for imaging of potential markers in tumors. In the practical part, the achieved published results are commented. Using MALDI-TOF MSI, the spatial distribution of estradiol 3,4 quinone, phytochelatins PC2 and PC3, oxidized glutathione (GSSG) and metallothioneins was determined in transverse sections of the Eisenia fetida. Furthermore, this method was used to map metallothionein in sections of prostate tumor murine xenografts (PC-3). In a case of monitoring the processes involved in the spontaneous regression of melanoma in MeLiM (Melanoma-bearing Libechov Minipigs) miniature pigs, this method was used to classify specific m/z peaks that significantly differentiated healthy skin and melanoma from MeLiM miniature pigs.
