Immunotoxicology of cyanobacterial metabolites

Abstract:

Masivní rozvoj cyanobakteriálního vodního květu představuje environmentální a zdravotní problém světového měřítka. Zatímco některé cyanobakteriální toxiny, jako např. peptidy microcystiny, byly intenzivně zkoumány, jiné toxické komponenty cyanobaketrií (např. lipopolysacharidy, LPS) byly jen částečně prostudovány. Předložená bakalářská práce sumarizuje v první části literární údaje o sinicových LPS a účincích LPS, stejně tak jako ostatních metabolitů sinic, na IS (údaje z experimentálních prací i epidemiologických studií). V praktické části je popsán experimentální postup izolace a purifikace LPS. Izolace byla provedena metodou fenolové extrakce (HPW), kde došlo k oddělení vodné fáze, která obsahovala lipopolysacharid od organické fáze s proteiny. K purifikaci izolovaného LPS byla použita dialýza a enzym RNáza. Byly izolovány LPS z 11 vzorků biomasy sinic z nádrží ČR dominovaných jedním z nejfrekventovanějších cyanobakteriálních rodů VKS (M. aeruginosa). Výtěžky LPS se pohybovaly od 0,4 do 1,5 mg z původní navážky 200mg sušiny. Endotoxinová aktivita (pyrogenita) purifikovaného LPS byla vyhodnocena LAL testem (Limulus amoebocyte Lysate test). Endotoxinová aktivita extraktů se pohybovala v rozmezí 746-2921 EU/mg biomasy.

Abstract:

Masive cyanobacterial water blooms are serious environmental and health problem worldwide. While some cyanobacterial toxins such as peptide microcystins were extensively investigated, other toxic compounds of cyanobacteria (e.g. lipopolysaccharides, LPS) are poorly understood. This bachelor thesis sumarizes in the first part available literature data about cyanobacterial LPS and effects of LPS as well as other cyanobacterial metabolites on immune system (data from experimental works and epidemiological studies). In the practical part of this thesis, experimental izolation and purification of LPS has been performed. The izolation was done by phenol extraction method, water phase with LPS was separated from the organic phase containing proteins. Extracts were purificated by dialysis and enzymatically using RNaze. LPS was isolated from 11 samples of cyanobacterial biomass from reservoirs in the Czech Republic (all samples dominated by one of the most frequently occuring cyanobacteria Microcystis aeruginosa). Content of LPS in the samples ranged from 0,4-1,5 mg (from original biomass dry weight 200 mg).The endotoxin activity (pyrogenicity) of purified LPS was measured by the LAL test (Limulus amoebocyte Lysate test). The endotoxin activities of cyanobacterial extracts ranged from 746-2921 EU/mg DW.
