Adaptation of plant metabolism to drought and production of compatible solutes

Abstract:

Rostliny během svého životního cyklu čelí různým změnám klimatu. Těmto změnám odolávají díky řadě buněčných i metabolických adaptací. Jednou z nich je produkce kompatibilních osmolytů, látek, které jsou hlavním tématem teoretické části práce. Zahrnuty jsou informace o jejich typech, charakterizaci, biosyntéze, katabolismu a biologické funkci. V teoretické části jsou také popsány metody stanovení těchto sloučenin. Experimentální část práce byla věnována vývoji a optimalizaci HILIC-ESI-MS metody kvantitativní analýzy kompatibilních osmolytů v rostlinných matricích. Důraz byl kladen na zvýšení citlivosti a zkrácení doby analýzy oproti doposud využívaným metodám. V rámci optimalizace byly rovněž porovnány dva různé extrakční postupy. Výsledná metoda byla 15 min dlouhá a u vybraných sloučenin poskytovala limity detekce na úrovni desítek fmol. Optimalizovaná metoda byla poté zvalidována a použita ke stanovení a porovnání obsahu kompatibilních osmolytů v semenech a prýtech hrachu setého. V semenech byly nejzastoupenějšími látkami rafinosové oligosacharidy, zejména stachyosa a verbaskosa, a také sacharosa. V prýtech byla nejabundantnějším metabolitem sacharosa. Nejméně zastoupeným metabolitem byl v obou matricích glycinbetain. Tyto výsledky odpovídaly odborné literatuře, nově vyvinutá metoda je tedy vylepšením doposud publikovaných postupů.

Abstract:

Plants face a number of climate changes during their life cycle. They restist them by a series of cellular and metabolic adaptations. One of these adaptations is production of compatible solutes, the substances which are the main topic of the theoretical part of the thesis. It includes the information about their types, characterization, biosynthesis, catabolism and biological function. The theoretical part also describes methods of determination of these compounds. The aim of the experimental part of this thesis was to develop and optimize a HILIC-ESI-MS method for quantitative analysis of compatible solutes in plant matrices. The optimization was done by increasing the sensitivity and shortening the total time of the analysis comparing to the previous methods. Another part of the optimization was to compare two different extraction protocols. The final method was 15 min long and provided detection limits of tens of fmol for selected compounds. The optimized method was then validated and used to determine and compare the content of the compatible solutes in pea seeds and shoots. Raffinose family oligosaccharides, especially stachyose and verbascose, and also sucrose were the most abundant metabolites in seeds. Sucrose was also the most abundant metabolite in leaves. Glycinebetaine was the least abundant metabolite in both matrices. These results corresponded to the literature, so the newly developed method is an improvement on previously published protocols.
