T cell marker characteristics for adoptive immunotherapy

Abstract:

The main goal of immunotherapy is the restoration and augmentation of the anti-tumor response, which was damaged or is under-developed. In practice, this type of treatment is used rather as an additive method in cases, where the main tumor mass has already been removed by another method. Thus immunotherapy should help to eliminate the residual disease and to ensure long-term effect (tumor remission). Adoptive transfer of T-lymphocytes, particularly cytotoxic T-lymphocytes, which are directly involved in the elimination of tumor cells, is a viable variant of immunotherapy. Helper T-lymphocytes also play an important role in the anti-tumor responses, because they activate other cells of the immune system and produce stimulative agents-cytokines. Both types of T-lymphocytes were identified in tumor tissue as tumor infiltrating lymphocytes (TIL). TIL for adoptive transfer are first isolated and subsequently expanded to clinically usable quantities. The partial success of this treatment procedure has been reported in cases of patients with melanoma, however, more efficient methods still need to be developed. The experimental part of this work dealt with the expansion of T lymphocytes in vitro by using three different media. Stimulated T-lymphocytes were isolated from peripheral blood of 3 healthy donors. The highest number of cells was achieved in the medium OpTmizer, which should be used for the stimulation of TIL in melanoma treatment project.

Abstract:

Hlavným cieľom imunoterapie je snaha o obnovenie porušenej imunitnej odpovede proti nádoru alebo posilnenie nedostatočne rozvinutej reakcie. V praxi sa tento druh liečby využíva skôr ako doplnková metóda v prípadoch, kedy hlavná nádorová masa bola odstránená iným spôsobom. Imunoterapia by tak mala prispieť k eliminácii zostatkovej choroby a zaručiť dlhodobé vyliečenie (remisiu nádoru). Možnou variantou imunoterapie je adoptívny transfer T-lymfocytov, predovšetkým cytotoxických T-lymfocytov, ktoré sa priamo podieľajú na eliminácií nádorových buniek. Dôležitú úlohu pri protinádorových reakciách zohrávajú i T pomocné lymfocyty aktiváciou ďalších buniek imunitného systému a produkciou stimulačných látok-cytokínov. Oba druhy týchto T-lymfocytov boli identifikované v nádorovom tkanive ako tumor infiltrujúce T-lymfocyty (TIL). TIL určené pre adoptívny transfer sú najskôr izolované a následne expandované na klinicky využiteľne množstvá. Čiastočné úspechy tohto liečebného postupu boli zaznamenané u pacientov s melanómom, avšak neustále je potrebné pracovať na ich zoptimalizovaní. Experimentálna časť tejto práce sa zaoberala expanziou T-lymfocytov in vitro s použitím 3 rôznych médií. Stimulované T-lymfocyty boli izolované z periférnej krvi 3 zdravých darcov. Najvyšší počet buniek bol dosiahnutý v médiu OpTmizer, ktoré by sa malo použiť i na stimuláciu TIL v projekte liečby melanómu.
