Determination of selected keto acids in biological samples using gas chromatography with mass spectrometry

Abstract:

Stanovení 2-oxokyselin má klinický význam, protože 2-oxokyseliny jsou klíčovými meziprodukty v řadě hlavních biochemických cest. U pacientů s některými dědičnými metabolickými chorobami jsou hladiny 2-oxokyselin zvýšené, např. u fenylketonurie. Tato práce se zabývá zavedením metody plynové chromatografie s hmotnostní detekcí pro stanovení 2-oxokyselin v biologických vzorcích. Metoda dokáže simultánně stanovit sedm významných 2-oxokyselin (pyruvát, 2 oxoglutarát, fenylpyruvát, 2-oxoisovalerát, 2-oxoisokapronát, 2-oxo-3-methylvalerát, oxalacetát). Za účelem derivatizace oxoskupiny 2-oxokyselin byly testovány tři derivatizační činidla, 2,4-dinitrofenylhydrazin, O-(2,3,4,5,6-pentafluorobenzyl)hydroxylamin a o fenylendiamin, derivatizace karboxylové skupiny pak N methyl N (trimethylsilyl)-trifluoroacetamid v 1% trimethylchlorsilanu, N,O bis(trimethylsilyl)trifluoroacetamid v 1% trimethylchlorsilanu, směs methanolu a acetylchloridu a BF3 v ethanolu. Pro separaci derivátů 2-oxokyselin byla použita kapilární kolona HP-5, kdy byly deriváty 2-oxokyselin separovány do 40 minut. Linearita byla dosažena v celém rozsahu testovaných koncentrací. Přesnost v sérii, vyjádřená variačním koeficientem, se u jednotlivých 2-oxokyselin výrazně lišila. Variační koeficient přesnosti stanovení pyruvátu a 2-oxoisokapronátu v plazmě byl do 7 %. Variační koeficient přesnosti stanovení dalších 2-oxokyselin byl i několikanásobně vyšší.

Abstract:

Determination of 2-keto acids is of clinical importance because 2-keto acids are key intermediates in a number of major biochemical ways. In patients with some inherited metabolic diseases, levels of 2-keto acids are increased, e.g. phenylketonuria. This work deals with the development of a gas chromatography with mass spectrometry for the determination of 2-keto acids in biological samples. The method can simultaneously identify seven significant 2-keto acids (pyruvate, 2-oxoglutarate, phenylpyruvate, 2-oxoisovalerate, 2-oxoisocaproate, 2-oxo-3-methylvalerate, oxalacetate). Three derivatizing agents, 2,4-dinitrophenylhydrazine, O (2,3,4,5,6 pentafluorobenzyl)hydroxylamide and o phenylenediamine, for a derivatization of carbonyl groups of 2-keto acids were tested. Derivatizing agents N-methyl-N-(trimethylsilyl)trifluoracetamide with 1% trimethylchlorosilane, N,O-bis(trimethylsilyl)trifluoroacetamide with 1% trimethylchlorosilane, a mixture of methanol and acetyl chloride and BF3 with ethanol, for derivatization of the carboxyl group of 2-keto acids, were tested. The HP 5 capillary column was used to separate the 2-keto acid derivatives within 40 minutes. The linearity was reached throughout the range of tested concentrations. The intra-assay, expressed by the coefficient of variation, differed significantly for the individual 2-keto acids. The variation coefficients for pyruvate and 2 oxoisocaproate in plasma were up to 7%. The variation coefficients for the other 2 keto acids were even several times higher.
