Metformin Deposition and its Antibacterial Properties

Abstract:

Metformin je perorální antidiabetikum, které se běžně používá pro léčbu diabetes mellitus 2. typu především u pacientů s nadváhou. V současné době však není metformin spojován pouze s diabetem jako antihyperglykemické léčivo. Zkoumají se jeho možné příznivé účinky při léčbě dalších onemocnění, jako je rakovina, kardiovaskulární a neurodegenerativní onemocnění, stejně tak jeho antibakteriální nebo antivirové účinky. Cílem této diplomové práce je depozice metforminu na povrch zvoleného syntetického polymerního materiálu, následná charakterizace připravených substrátů dostupnými metodami a zjistit, zda má metformin potenciál jakožto antimikrobiální činidlo. Nejprve byl metformin deponován pomocí vícestupňového fyzikálně-chemického procesu na polymerní nosič z nízkohustotního polyethylenu (LDPE). Tento proces zahrnoval ošetření povrchu pomocí plazmového výboje, navázání kyseliny akrylové na povrch a EDC, který umožnil navázání aminových skupin metforminu s povrchovými karboxylovými skupinami. Připravené substráty byly charakterizovány pomocí FTIR, měření mezního kontaktního úhlu a povrchové energie, SEM a XPS. K determinaci antibakteriální aktivity připravených povrchů s deponovaným metforminem byly použity kmeny bakterií Escherichia coli (CCM 4517) a Staphylococcus aureus (CCM 4516), přičemž samotné testování probíhalo podle norem ČSN ISO 22196 a ČSN EN ISO 20743. Na základě použitým metod bylo prokázáno, že depozice metforminu na povrch LDPE proběhla úspěšně. Stejné tvrzení ovšem neplatí v případě výsledků z antibakteriálního testu. Z toho totiž vyplývá, že metformin prokazuje pouze minimální antibakteriální aktivitu, vůči použitým bakteriálním kmenům. Jediný vzorek, u kterého se naměřila antibakteriální aktivita byl vzorek s koncentrací deponovaného metforminu 1 gl-1 vůči bakterii Escherichia coli. Ze získaných výsledků se dá tedy konstatovat, že využití metforminu jakožto antibakteriálního činidla se zdá být velmi omezené. Nicméně v době odevzdání této práce nebyly k dispozici výsledky z antibakteriálního testu vzorků s vyšší koncentrací deponovaného metforminu a budou tak předmětem dalšího zkoumání.

Abstract:

Metformin is an oral antidiabetic drug that is commonly used for treating type 2 diabetes mellitus, especially in overweight patients. However, nowadays, metformin is no longer associated only with diabetes as an antihyperglycemic drug. Its possible beneficial effects in the treatment of other diseases such as cancer, cardiovascular and neurodegenerative diseases, as well as its antibacterial or antiviral effects are being investigated. The aim of this diploma thesis was to deposit metformin on the surface of a selected synthetic polymer material, further characterize the prepared substrate using available methods and determine whether metformin has potential as an antimicrobial agent. Firstly, metformin was deposited using physico-chemical multistep process onto a polymeric carrier from low-density polyethylen (LDPE). This process involved treating the surface using plasma, bonding of acrylic acid to the surface and EDC, which binds the surface carboxyl groups with metformin amine groups. The prepared substrates were characterized by FTIR, measuring the limiting contact angle and surface energy, SEM and XPS. The bacterial strains of Escherichia coli and Staphylococcus aureus were used to determine the antibacterial activity of the prepared surfaces with deposited metformin, and the testing itself was carried out according to ISO 22196 and ISO 20743 standards. Based on the used methods, it was shown that the deposition of metformin on the LDPE surface was successful. However, the same statement is not true for the results from the antibacterial test. This determined that metformin shows only minimal antibacterial activity against the bacterial strains used. The only sample for which antibacterial activity was recorded was a sample with a deposited metformin concentration of 1 gl-1 against Escherichia coli. Thus, from the results obtained, it can be concluded that the use of metformin as an antibacterial agent appears to be very limited. However, at the time of submission of this thesis, results from the antibacterial test of samples with higher concentrations of deposited metformin were not available and thus will be subject of further investigation.
