Prediction of the probability of the pathogen occurrence in populations of rodents

Abstract:

Populace drobných savců žijících na území ČR jsou významnými rezervoáry některých patogenních zoonotických onemocnění. Významný vliv na míru výskytu daných patogenů v populacích hlodavců můžou mít klimatické i jiné podmínky. Cílem této práce je odhalit takové proměnné prostředí, které statisticky významně ovlivňují výskyt vybraných patogenů a využít je k sestavení modelů pro predikci výskytu infekčních agens v populacích drobných savců. Potřebná data byla získána terénním odchytem drobných savců a sledováním proti nim vytvořených protilátek v séru a výplachu srdce. Odchyty byly prováděny v letech 2010 – 2014. Celkem bylo odchyceno 679 jedinců druhů Apodemus flavicollis, A. agrarius, A. sylvaticus, Clethryonomys glareolus, Microtus arvalis a Sorex araneus. Metodou ELISA byla detekována přítomnost specifických IgM a IgG protilátek proti Borrelia burdorferi s.l., Coxiella burnetii a Francisella tularensis. Pomocí metody MAL byly zjišťovány celkové protilátky proti Leptospira interrogans s.l.. K nalezení vhodného modelu byla použita logistická regrese, která umožňuje predikovat binární proměnné. Z výsledků sestavených predikčních modelů vyplývá, že výskyt Bbsl pozitivně ovlivňují počty mrazových a ledových dnů spojených s nízkou sněhovou pokrývkou v zimě před odchytem, negativní vliv má průměrný tlak vodní páry. Menší šanci na nakažení C. burnetii a F. tularensis mají samci. Výskyt C. burnetii pozitivně ovlivňuje počet letních dní v roce před odchytem i počet ledových dnů v zimě před odchytem. Pravděpodobnost výskytu F. tularensis je významně vyšší na jaře, než v létě nebo na podzim, pozitivně ji ovlivňuje i teplota na podzim a v létě před odchytem a počet dní se srážkami alespoň 1mm v měsíci. Na výskyt Lisl pozitivně působí průměrná vlhkost vzduchu, ve spojení s nízkým tlakem v měsíci odchytu. Výsledné modely lze použít pro predikci výskytu patogenů v populacích drobných savců a pro prevenci přenosu infekčního agens na člověka.

Abstract:

Populations of small mammals living in the Czech Republic are major reservoirs of certain pathogenic zoonotic diseases. Climatic and environmental conditions can have significant influence on the prevalence of the pathogens in the rodent population. The aim of this work is to identify those environmental variables that have significant affect on the incidence of selected pathogens and use them to build predictive models of the occurrence of infectious agents in small mammal population. Necessary data were obtained by trapping small mammals and by observation of antibodies against these pathogens in their serum and heart rinses. Trapping were conducted in the years 2010 - 2014. 679 mammals of species Apodemus flavicollis, A. agrarius, A. sylvaticus, Clethryonomys glareolus, Microtus arvalis and Sorex araneus were caught. ELISA was used to detect the presence of specific IgM and IgG antibodies against Borrelia burdorferi s.l., Coxiella burnetii and Francisella tularensis. Method MAL was used to detect the total antibodies against Leptospira interrogans s.l.. Logistic regression for prediction binary variables was used to find a suitable model. Number of frost and ice days associated with low snow cover in the winter before trapping affects positively incidence of Bbsl, negative influence on Bbsl incidence has average water vapor pressure. Males have less chance to infection by C. burnetii and F. tularensis. Number of summer days in the year before trapping and number of ice days in the winter before trapping have positive influence on the incidence of C. burnetii. Probability of F. tularensis is significantly higher in the spring than in the summer or fall. Temperature in the autumn and in the summer before trapping and number of days with precipitation at least 1 mm in the trapping month affect positively the incidence of F. tularensis. Average air humidity together with low pressure has positive influence on the occurrence of Lisl. The final models can be used to predict the occurrence of pathogens in small mammal populations and for preventing transmission of infectious agents to humans.
