The analysis of microRNA expression and function in the pathogenesis of lymphomas and leukemias

Abstract:

MicroRNA (miRNA) jsou důležité post-transkripční regulátory genové exprese a jejich deregulace hraje důležitou roli v patogenezi a prognóze hematologických malignit včetně folikulárního lymfomu (FL). FL je nejčastějším indolentním lymfomem s velmi variabilním klinickým průběhem a nepříznivou událostí v jeho vývoji je histologická transformace do agresivnějšího typu lymfomu, nejčastěji do difuzního velkobuněčného B-lymfomu (DLBCL). V posledních letech se ukazuje, že s transformovaným FL (tFL) souvisí aberace a zvýšená exprese onkogenu MYC, nicméně přesné molekulární mechanismy tohoto procesu jsou doposud nejasné. V této disertační práci bylo provedeno první globální profilování exprese miRNA v párových vzorcích FL a tFL a bylo identifikováno pět odlišně exprimovaných miRNA. Práce se podrobně zaměřuje především na miR-150, jejíž exprese ve všech zkoumaných tFL uniformně klesá. Ukázalo se, že tento pokles souvisí s vysokými hladinami proteinu MYC, který v B buňkách potlačuje expresi miR-150 přímou vazbou do její DNA. V práci je dále ukázáno, že nízké hladiny miR-150 v tFL vedou ke zvýšené produkci jejího cílového proteinu FOXP1, který je známým pozitivním regulátorem přežívání buněk a signalizace přes BCR a NF-κB v maligních B buňkách. Dále bylo popsáno, že nízké hladiny miR-150 a vysoké hladiny FOXP1 asociují s kratším přežitím pacientů s FL. Kvantifikace exprese miR-150 by tak mohla sloužit jako biomarker snadno detekovatelný v tkáni fixované formalinem a zalité v parafinových blocích (FFPE tkáň). Celkově tato studie prokázala významnou roli dráhy MYC/miR-150/FOXP1 v nádorových B buňkách v souvislosti s agresivitou a transformací FL.

Abstract:

MicroRNAs (miRNAs) are important post-transcriptional regulators of gene expression and their deregulation plays an important role in the pathogenesis and prognosis of hematologic malignancies including follicular lymphoma (FL). FL is the most common indolent lymphoma with a very variable clinical course and the adverse event in its development is a histological transformation to a more aggressive type of lymphoma, typically to diffuse large B-cell lymphoma (DLBCL). Recent studies show that the transformed FL (tFL) is associated with aberrations and overexpression of MYC oncogene, but the exact molecular mechanisms of this process are unclear. In this thesis, we performed the first global profiling of miRNA expression in paired FL and tFL samples, which identified five differentially expressed miRNAs. The work focuses particularly on miR-150, whose expression decreases uniformly in all studied tFL cases. This miR-150 decrease has been shown to be related to high levels of MYC protein, which suppresses miR-150 expression in B cells by direct binding in its upstream region in DNA. We further demonstrated that low levels of miR-150 in tFL result in increased production of its target protein, namely FOXP1, which is a known positive regulator of cell survival as well as BCR and NF-κB signaling in malignant B cells. Furthermore, this study reported that low levels of miR-150 and high levels of FOXP1 associate with shorter survival of FL patients, which suggests that miR-150 expression could serve as a good biomarker readily detectable in formalin-fixed paraffin-embedded (FFPE) tissue. Overall, this study demonstrated an important role of the MYC/miR-150/FOXP1 axis in tumor B cells as a determinant of FL aggressiveness and high-grade transformation.
